This is without a doubt the most brilliant and poignant depiction of HPLC and CE of biomolecules in modern history. My extreme hyperactivity disorder usually means that I'm not able to sit still for such weighty topics--not without large amounts of abused Ritalin; but Cunico's work was so compelling and finely wrought that I found myself spellbound, even when drunk, even when driving in the backseat of a stolen car across the Mexican border at Tecate. I am not going to recommend drinking heavily and stealing cars. But I do recommend BASIC HPLC and CE OF BIOMOLECULES by Robert L. Cunico. By the way, I am extremely drunk right now.
Basic HPLC and CE of Biomolecules.......2000-12-24
This is a book that should be on the desk of anyone who is involved in analytical development of biomolecules and biopharmaceuticals. The content is well organized and written in a format that is easy to grasp and understand. The content covers almost all the different modalities of HPLC analyses and applications and provides real life examples of the techniques. It includes innumerable suggestions on how best to develop a validatable HPLC method and analyze the data appropriately.
I would highly recommend this book to both new and old members of biotechnology, especially the developmental, analytical, and quality assurance groups where there is frequently a need to implement a solid reliable method for analyzing biomolecules in a more rational fashion.

Description:
Capillary array electrophoresis (CAE) is an important high throughput analytical technique. Laser-induced fluorescence (LIF) has been the dominant detection method for CAE owing to its low limit of detection (LOD) and wide linear dynamic range (LDR). Linear LIF scanners were first used in CAE because linear motions of an objective match well with a common planar array of capillaries. A problem with linear scanners is that the motor is required accelerating/decelerating so that all capillaries can be properly scanned, which makes motion control complicated and reduces the duty cycle. Rotary scanners were developed to overcome this problem. While rotary scanners have been successfully applied in CAE, the capillaries have to be arranged in a circular format, which can be inconvenient in some cases. In this report, we describe a cam-based LIF scanner as an alternative technique for CAE detection. In this system, a rotary motor is mechanically linked with a capillary holder via a cam. During operation, the motor carries the cam in a rotary motion that drives an array of capillaries on the holder to move back and forth across the objective for fluorescence detection. Using this design, the capillaries can be parallel-arranged in a plane while the motor acceleration/deceleration is avoided. To demonstrate the feasibility of this approach, we constructed a prototype instrument with a constant-velocity scanning distance of ~10mm, a scanning frequency of 3Hz and a duty cycle of ~70%. The scanner exhibited a LOD of 69pM of fluorescein and a LDR of 3.5 orders of magnitude. Multiplexed capillary SDS-PAGE was performed on this scanner for protein separations.

Description:
A highly sensitive nanomechanical cantilever sensor assay based on an electrical measurement has been developed for detecting activated cyclic adenosine monophosphate (cyclic AMP)-dependent protein kinase (PKA). Employing a peptide derived from the heat-stable protein kinase inhibitor (PKI), a magnetic bead system was first selected as a vehicle to immobilize the PKI-(5-24) peptide for capturing PKA catalytic subunit and the activity assay was applied for indirectly assessing the binding. Synergistic interactions of adenosine triphosphate (ATP) and the peptide inhibitor with the kinase were then investigated by a solution phase capillary electrophoretic assay, and by surface plasmon resonance technology which involved immobilization of the peptide inhibitor. After systemically evaluated by a homogeneous direct binding assay, the ATP-dependent recognition of the catalytic subunit of PKA by PKI-(5-24) was successfully transferred on to the nanomechanical cantilevers at protein concentrations of 6.6pM-66nM, exhibiting much higher sensitivity and wider dynamic range than the conventional activity assay. Thus, direct assessment of activated kinases using the cantilever sensor system functionalized with specific peptide inhibitors holds great promise in analytical applications and clinical medicine.

In the present work, protein digest reactions in silicon-based microchips, coated with immobilized proteolytic enzymes, have been carried out. The performance of such vials, modified with trypsin or chymotrypsin, was tested with myoglobin as a substrate. Capillary electrophoresis and matrix-assisted laser desorption/ionization mass spectrometry were utilized for analysis of the digests, and the influence of different instrumentation setups, immobilization procedures and reaction conditions are discussed.

Call it Robert Fulghum by way of John Wayne... In 1953, Zell Miller was as low as he could get. He'd dropped out of college after being made to feel inferior because of his "hillbilly" background and wound up in jail one weekend after getting drunk on moonshine and driving his car into a ditch. In an effort to turn his life around, he signed up for marine boot camp. The experience changed his life, and he remains convinced that the values he learned during his 90 days at Parris Island are "the only basis upon which diversity can coexist with commonality and all people can pursue individual goals for themselves while contributing to the general well-being and advancement of society as a whole." These simple values, from neatness and punctuality to discipline and loyalty, are for Miller the basis of a strong civil society. Although some readers may find some of his notions--such as his frustration at seeing kids wear caps backwards--a bit extreme, Miller reminds us that any organization that pumped out men like Bernard Shaw, Don Imus, Ted Williams, and Art Buchwald must have something going for it.

I agree with LTC Kennedy. This is a great book for instructing young people. I do wish that Gov. Miller would read it for books-on-tape, most certainly in an unabridged version. Some of us just learn so much more listening to the voice and twang of a good Southern teacher, much like Shelby Foote.

Recently while conducting a manager orientation I realized that every value we want our managers to hold is expressed in Corps Values.
Zell Miller captures the essence of what is missing in our approach to our everyday dealings.Anyone who has read The 7 Habits of Highly Effective People should also read Caorps Values. They go hand-in-hand.
I have purchased copies for all of our managers and plan on having my children read this once they hit their teen years.Corps Values is a great resource for anyone looking for direction. This is true self-help.

Zell learned a great deal from the Marines. Honesty, loyalty, personal pride, respect. The Marines are not the only place these things are taught, but it is one of the finest. So many young people today have never been taught any of these. It is a shame that many of our youngest (and possibly, our brightest) are doomed to failure in life because of the things parents and society SHOULD have taught them. Fortunately, the Marines haven't changed.

This book is an easy delight to read. Anyone who has ever had a passion for life, other than our daily dispositions, will covet the easy-to-grasp concepts about 'core values.' However, we all know, try with all our might, these concepts (virtues) are not that simple to master. It takes a lifetime of knowledge, information and experiences for us to bear the fruits of our existence. It takes a consistent flow of discipline!
First, I want to heartily thank the honorable Zell Miller for his generous lust for life! All of my life, I have been searching for a mentor who I could respect with the brightest colors of honor, dignity, courageousness and passion. I read about them, but have never heard or seen one! (Boy would I love to meet him!!!)
When I saw Zell Miller, as a Dixie Southern Democrat from Georgia, speak with a twang at the Republican National Convention on television; I could not hold the tears away from my eyes. I witnessed the greatest oratory in my life. The mastery and delivery of his speech put all the other presentations in the dump in comparison to his feisty 'rooster-calls.' 'Moral authority' and all his `core values' were oozing from the impeccable Zell Miller. I recognized in a heartbeat how quickly I would pick up my pistol (former Navy Corpsman, Reserves, and proudly attached to Marine Corps, Reserves) at his calling and fight with him to hell and back!
This book is an affirmation of my 'corp beliefs.' It is a refresher course for virtues that almost disappeared due to strong influences from everyone else.
America is reflected as a promise for a free society. A free society demands [broad'er] "characteristics which constitute the difference between a responsible and contributing human being and an unconscionable savage bent upon taking or destroying whatever they want that they are unable or unwilling to earn for themselves." (Miller, p. 105)
We should test our liberalism and our conservatism, as long as the human condition does not sway to anarchy and we do not get 'self-absorbed' by too many excesses in any direction. This is America's challenge and that, my friends, is our Birthright.

Three decades after his death, the life and career of Supreme Court Justice Hugo L. Black continue to be studied and discussed. This latest and perhaps definitive study of Black's origins and early influences has been 25 years in the making and offers fresh insights into the justice's character, thought processes, and instincts. Black came out of hardscrabble Alabama hill country, and he never forgot his origins. He was further shaped in the early 20th-century politics of Birmingham, where he set up a law practice and began his political career, eventually rising to the U.S. Senate, from which he was selected by FDR for the high court. Black's nomination was opposed partly on the grounds that he had been a member of the Ku Klux Klan. One of the book's conclusions that is sure to be controversial is that in the context of Birmingham in the early 1920s, Black's joining of the KKK was a progressive act. This startling assertion is supported by an examination of the conflict that was then raging in Birmingham between the Big Mule industrialists and the blue-collar labor unions. Black, of course, went on to become a staunch judicial advocate of free speech and civil rights, thus making him one of the figures most vilified by the KKK and other white supremacists in the 1950s and 1960s.

Response time was very short. You couldl tell that the book was used, but nothing beyond what would be considered "normal" wear. Overall conditioin was very good.
A superbly researched and written biography of Hugo Black.......2005-10-11
This superbly researched and written biography of Hugo Black recreates for the reader the times in which the Deep South was bound up by traditions of white supremacy and how a Southern white man developed a judicial philosophy and temperament to help end America's legal segregation and restore a simple justice that was the hoped for outcome of the American civil war, but which had been undercut by the development of a "Jim Crow" social order of repression and segregation. Biographer Steve Suitts provides new and pivotal information as he lays out the story of Black's personal and public life, provides new perspectives on the sweeping forces that shaped the destiny of Black's life, and the struggle for racial justice in the first quarter of the 20th century. A work of impressive and accessible scholarship, Hugo Black Of Alabama is a highly commended addition to community and academic library American Biography and Judicial History collections.

Very timely, with the recent death of Judge Rehnquist, the book gives and in-depth picture of a man who follows his own ideals of truth, justice and the equality of all men, regardless of color or faith.
IMHO Suitts's is a third rate hack.......2005-07-21
I've only read bits and pieces of his book, but a recent opinion piece that he penned in the Fulton County Daily Report--in which he maliciously attacks a sitting federal judge and attempts to whitewash Hugo Black's vile racist/anti-Catholic tactics during an infamous Birmingham, Alabama murder trial in the 1920s--strongly suggests to me that Suitts's book is not worth the trouble.
In a word, Suitts appears to me to be nothing more than a liberal, partisan hack.
