The molecular biology revolution has required the development of new chromatographic techniques and the optimization of original techniques to give reasonable quantities of protein at high resolutions. The aim of this volume is to provide the necessary information in most experimental situations to enable rapid and effective purification. The first four chapters deal with the instrumental aspects of high resolution chromatography starting with the initial clean up steps prior to separation in chapter 1. Chapter 2 deals with microscale techniques, then chapter 3 describes the detector technologies that can determine information about the separated molecules. The final chapter in this section cover capillary electrophoresis and its associated techniques. The remaining chapters cover a range of chromatographic procedures based on the interaction of a specific ligand with its target protein or other macromolecule. Some chapters cover non-specific interactions using peptides, inhibitors, and antibodies as the affinity ligand while others focus on specific groups of molecules : oligosaccharides and glycosylated proteins, nucleotide-binding proteins, proteins binding free and chelated metal ions, and DNA binding proteins.

Una, named for the heroine of Edmund Spenser's Faerie Queene, flees to the New England coast from Kentucky to escape her father's puritanism and to pursue a more exalted life. She gets whaling out of her system early: going to sea at 16 disguised as a boy, Una has her ship sunk by her own monstrous whale, and survives a harrowing shipwreck:
I was so horrified by the whale's deliberate charge that I could not move. Then my own name flew up from below like a spear: "Una!" Giles' voice broke my trance, and I scrambled down the rigging. No sooner did my foot touch the deck than there was such a lurch that I fell to my face. I heard and felt the boards break below the waterline, the copper sheathing nothing but decorative foil. The whole ship shuddered. A death throe.
The ship dies, but Una returns to land to pursue the life of the mind. The novel's opening line--"Captain Ahab was neither my first husband nor my last"--also diminishes Melville's hero in the broader scheme of things. Naslund exposes the reader to the unsung, real-life heroes of Melville's world, including Margaret Fuller and her Boston salon, and Nantucket astronomer Maria Mitchell. There is a chance meeting with a veiled Nathaniel Hawthorne in the woods, and throughout the novel the story brims with references to the giants of literature: Shakespeare, Goethe, Coleridge, Keats, and Wordsworth. Ahab's Fictional Wife's Philosphical Musings and Deep Inner Life is a title that aptly describes this endless novel. The segments that were plot-driven, such as Una's adventures as a sailor and the mental illness of her first husband, were wonderful. Ahab, until now merely an icon, was infused with genuine humanity, and the story of their marriage was so well done. Life on Nantucket and the experience of sea captains' wives are vividly drawn. Naslund is a very good writer. It was all the stuff in between (Una's odes to her sewing needle, her effusive love affair with the seas, her genius despite her poor education, to offer just a few out of dozens of examples) that brings my rating down to 3 stars. Maybe this new writer was given a new editor. Someone should have blue pencilled about 200 pages of filler. Certainly Abundance is a much tauter production. This book might have been equally good.

Ahab's Wife serves admirably as a companion book to Melville's Moby-Dick and having read both, I think I can safely say that if Herman Melville were to read Ahab's Wife, he would be more than happy with the duty and accuracy Naslund devotes to the period, the prose, and its homage to Melville's opus.
This is the life story of Una, the wife of Ahab - the peg-legged determined-bordering-on-insane captain of the Pequod in search of his white whale. The cover of the book depicts a Puritan-clothed woman on a harsh beach looking out into a rough sea, while further down the beach lies the broken hulk of an old ship. It creates images and ideas of a worrying woman left at home for years at a time to tend to house and children, while her husband is out braving the sea, fighting giants monsters in his man's world. One would think this a book about her everyday actions, her chores, her repetitive characteristics, and while this is part of the book, there is so much more going on in Una's life with her triumphs and tribulations, her loves and deaths, her dangerous adventures, and her happy times at home. This is what makes Ahab's Wife a welcome companion to Moby-Dick, for while Ahab's is a story of adventure and danger, Una's is just as much so.
The book begins, as all life stories should, with a birth, only Una's mother is all alone in a cabin and naturally it is a birth that almost kills her. Una's life is a harsh one in Kentucky and before she is ten, her mother sends her away to her aunt's. Una's father is a devout Christian, while Una is an atheist from a young age, choosing not to blindly believe in what her father tells her to believe. Her mother fearing for her life, sends her to the distant coast of New England to live with her aunt and uncle in a lighthouse. And so begins the next chapter in her life, with a different family, in a different place. With the arrival of two men who come to upgrade the lighthouse, she falls in love with both of them - even though she is still young - knowing that one will be her husband one day. At the age of eighteen, she leaves the island and the lighthouse for the mainland of Boston and then Nantucket getting by on simple work until she finds the same two men whom she loves on a whaling ship. Disguising herself as a young boy she joins the crew and experiences the whaling life of her future husband. It is here that she first sees The Pequod and meets Ahab, who by then is an old man but still respectable and honorable. Ahab is the one to marry Una to Kit when her existence on the ship, love for that man, and her femininity are all revealed.
A whale stoves in the ship and Una spends many days on a small boat with the remaining crew reduced to cannibalism - harking to the story of Moby-Dick as well as the story of the whale ship Essex, which was the impetus for Melville's story. It is on the return journey to Nantucket that the other love of her life dies tragically and her husband Kit essentially goes insane. Upon returning to land and leaving her husband due to his condition, Una's life slows down and her relationship with Ahab begins until their marriage and happiness together. It is here that the story of Moby-Dick truly begins and the reader gets to meet the familiar characters of the classic book. But while Ahab spends years away from home, Una's life goes on with the birth of a child and the struggles of her life. It is upon the return and meeting of Ishmael that Una learns of the doomed story of Ahab, his white whale, and his death.
The book could be considered technically over at this point, but this is the story of Una, who is still very much alive. The rest of her life is spent interacting with Ishmael and even meeting and interacting with the slave who fought for his freedom, Frederick Douglass. And while she never forgets her life with Ahab, she eventually finds another husband and in the waning years of her life is happy once more.
What makes Ahab's Wife a truly impressive book is not just its intended mimicry with Moby-Dick with the crossing over characters, similar layout of the book with many chapters and illustrations, and actual scenes involving the same location in both books such as the church with the pulpit carved to imitate the bow of a ship which the same preacher from Moby-Dick climbs the ladder to the top and scream of hellfire and damnation; it is the prose and how Naslund writes that truly emulates the style of Melville, making this a truly important work of literature deserving a place in the shelves with Melville, James and Hawthorne.

What awesome descriptions and captivating storyline! I couldn't put this book down! It makes me want to read "Moby Dick" again! I look forward to reading Sena Jeter Naslund's other novels. If they are half as good as the story of Una, I am hooked! I found that Una is so much like my daughter, and the author never met my daughter. Of course, this made the story even more interesting to me. The personalities are so diverse and very believable. I would recommend this book to everyone.

Awesome!.......2007-08-27
Love this book that I'm reading. I've never completed reading Jeremiah straight from the Bible cos I find it very heavy stuff. But after reading Run with The Horses, I'm beginning to appreciate this weeping prophet. I'm still mulling over those chapters that I've read and look forward to turning the pages still. Very sobering stuff in there for personal reflection. You can't ignore what you see/realise in yourself.
Outstanding Challenge to a Life of Excellence.......2007-06-05

"Run With the Horses" is not so much a commentary on the book of Jeremiah as it is a meditation on the life of Jeremiah. Though Peterson does move us chronologically through Jeremiah's life, he chooses only certain "episodes" or "situations" to discuss. Peterson ties his meditations on the life of Jeremiah to our lives as Christians, covering such topics as:
* Our identities as human beings with a definitive purpose.
* How we can be misled through deceptive teachings and words.
* How our choices can twart God's purposes for us.
* The importance of being honest and vulnerable before God.
* The significance of persistence and perseverance when enduring life's challenges.
* How God's ways and purposes often seem to contradict what we see and know from the world.
In essence, "Run With the Horses" is a meditation on the anatomy of a life of faith--a risky endeavor that is not for the faint of heart. Personally, I was more challenged and encouraged by this book than any other book I have read in recent memory. Highly recommended.

Never knew who Jeremiah really was.......2006-01-26
This book is a masterpiece. I have read the book of Jeremiah several times, but Peterson makes this prophet of old come alive. We end up feeling what Jeremiah felt; we see what he saw; we understand his fear. Peterson's intrepretation makes this Old Testament book apply personally to our lives whereas sometimes a person can read the Old Testament and have trouble seeing how it relates to modern day. God has surely blessed Peterson with gifts of writing, prophecy, interpretation and understand. Thank you Mr. Peterson for sharing your gifts with us all.
LIKE JEREMIAH, WE, TOO, CAN RUN WITH THE HORSES.......2005-09-17
God said to Jeremiah, "If you have raced with men on foot, and they have wearied you, how will you compete with horses?" To put it in modern language, "If you can't pay your bills on time, how are you going to walk on water?" Okay, sort of modern. The point is, God wants us to walk on water, and we're still struggling to pay our bills on time. That was Jeremiah's problem, until he realized, "No, I won't be able to do that in my own strength; but I will in Yours." And that was when Jeremiah changed from being a spiritual workhorse to being a thoroughbred.
No one says it like Eugene Peterson. He's one of the most eloquent writers around, and what he says is loaded with meaning. In this case, no one says what's involved in the quest for life at it's best like he does. In chapter 8, "My Wound Incurable," in Run with the Horses, he describes a picture of prayer that looks very much like having dinner in a fine restaurant with the most important person in your life. It's private, intimate, conversational, meaningful, personal - in a word, special. Occasionally, the waiter shows up to take your order, bring your food or take the broccoli back because it was cold; but otherwise, it's just you and your special loved one.
Then he writes, "But there is a parody of prayer that we engage in all too often. The details are the same but with two differences: the person across the table is Self and the waiter is God. This waiter-God is essential but peripheral. You can't have the dinner without him, but he is not an intimate participant in it. He is someone to whom you give orders, make complaints, and maybe, at the end, give thanks. The person you are absorbed in is Self - your moods, your ideas, your interests, your satisfactions or lack of them. When you leave the restaurant you forget about the waiter until the next time. If it is a place to which you go regularly, you might even remember his name.

But Jeremiah wasn't always like that. He had to start at square one like the rest of us. That is where he was when God asked, "What is it you really want, Jeremiah, do you want to shuffle along with the crowd, or run with the horses?" "The response when it came was not verbal, but biographical. His life became his answer, 'I'll run with the horses.'" So can we.

A Great Study Guide.......2007-05-12
I am blessed by this book because each study is simple and direct. There isn't a lot of reading in each section which keeps it easy for study groups to complete the assigned sections. The questions direct your attention to meanings you might have missed within the assigned verses of Jeremiah which can be a difficult read. Overall, I would suggest this book for any bible study group or individual.

Run with the Wind by Caroline Pitcher, Illustrated by Jane Chapman, is one o the best books I've ever read. It's just so cute. It has the most gorgeous illustrations that I have ever seen! This book is put together very well. Its main point and message tells of a mother and child's bond. It's a really good book for reading to your children. It really will let them know how much you love them and care about them. It's about a mother horse who has a foal. Sha tells him all the time of how much she loves him and how even though he can't see her doesn't mean that she's not with him. Just like the wind. He keeps telling her that he doesn't want to think about it and when the time comes for her to leave him, while she works, he has so much fun playing with the wind that he doesn't even think about her. This is a really good book and I highly recamend it.

Runs With Horses had always dreamed of becoming an Apache warrior, but knew of the numerous trials he would have to pass before he became one. Ever since he could walk, Runs With Horses had trained to become a warrior, and fight the Mexicans and White Eyes that tried to put his people into reservations, like his father, Red Knife, and his tribe leader, Geronimo. To prove him self he had to pass many tests to go on a raid. After completing four raids he would be considered a warrior, and eligible to smoke, drink, and marry. His tests ranged from running to the most distant mountains and back without being seen to dodging rocks slung at him by his father. Shortly after Runs With Horses went on his third raid, with had failed, Geronimo decided to surrender to the White Eyes and go to the reservations to be with his people. Runs With Horses never became a warrior, and was now trained to live like a white. Although this book had plenty of action, it was too close together in the story line to keep my attention. It was worth reading because of everything I learned from it, but it could have been better.
Many of Runs With Horses' quests were very exciting, and fun to read. On one of his missions, Runs With Horses had to sneak into the distant mountains to get information from a scout, and return back to his tribe undetected. While on his trip he spotted a small group of Mexicans camping in the valleys. He waited for them to fall asleep and planned to steal their three mules, donkey, food, and two of their rifles. After untied the animals and grabbing one rifle, one of the men woke up. He shot at the boy and scared of the animals in the process. Runs With Horses had to run all night before he knew he was a safe distance from the Mexicans. Exciting scenes like this are what kept me reading this book.
Another thing I liked was Runs With Horses' desire to succeed. When Red Knife told Runs With Horses that a lot of the tribe was surrendering, Runs With Horses replied, "Geronimo is not a coward! He will not give up. He will fight until the end." Runs with Horses had a lot of faith in his tribes ability and believed that they could defeat the Mexicans and White Eyes that "are more than the stars."
This book's biggest downfall is the amount of action it tries to shove into its pages. Good suspense stories have action then rest then have action again, but not this book. From cover to cover this book was action. If the author would have written about the time in between Runs With Horses' trials then it probably wouldn't have been as bad, but instead they went straight from trial to trial. The action was great but I would have enjoyed it more in moderation than all at one time.
Runs With Horses is a good book because of its educational value, excitement, and the characters. I would like to see a sequel so that I know what happens to Runs With Horses after he is trained to live like the whites, but it is unlikely that will happen. I recommend this book to anyone who is interested in Native Americans or just wants an exciting book to read.
