Book Description

Comprehensive, up-to-date coverage of a revolutionary technique Electrospray ionization mass spectrometry (ESI-MS) has completely changed the way physicists, chemists, and biologists view the study of large molecules. The technique derives detailed information about molecular weights and structures from extremely small sample quantities. ESI-MS can create highly charged forms of very high molecular weight compounds, it is naturally compatible with many types of separation techniques, and it allows noncovalent interactions between molecules in solution to be preserved in the gas phase. But many researchers may not use the technique to its full potential because they are unfamiliar with the different perspectives of its underlying processes, the varied approaches to implementation, and the wide-ranging utility of the technique.

In this book, Richard B. Cole and an assemblage of leading researchers present a single-volume compilation of different approaches to the understanding and exploitation of ESI-MS. This comprehensive guide:
* Examines the physical and chemical aspects of the electrospray process and describes the events involved in ion formation as well as the electro-chemical phenomena that are central to charged droplet formation during the process
* Explores the coupling of electrospray ionization to various mass spectrometers, including quadrupole, magnetic, time-of-flight, quadrupole ion trap, and Fourier transform ion cyclotron resonance instruments
* Describes recent progress in interfacing ESI with solution-based separation techniques, including liquid chromatography and capillary electrophoresis
* Charts the rapid development of ESI applications and categorizes them by compound type: peptides and proteins, nucleic acids and their constituents, carbohydrates and lipids, small molecules related to pharmacology and drug metabolism, and organometallics and inorganic compounds

Electrospray Ionization Mass Spectrometry is the indispensable handbook and reference for anyone who wishes to understand, implement, or apply this technique, including researchers in chemistry, metallochemistry, biochemistry, biology, pharmacology, and physics.

        Description:
Three procyanidin fractions, one with shorter oligomers, one with longer oligomers and one with polymers, were obtained from Pinus sylvestris L. bark extract. Procyanidins were analyzed using reversed-phase and normal-phase high-performance liquid chromatography methods coupled with electrospray ionization mass spectrometric detection in the negative ion mode. On the grounds of mass spectral data, pine bark was found to contain procyanidins from monomers through decamers and higher polymers.

          Description:
Soybean lecithin and phenol intensely interfered in the determination of insulin with HPLC-UV in an emulsion system which was developed for transmucosal delivery. Thus, in this study electrospray ionization mass spectrometry (ESI-MS) with ion trap detection was applied to screen extractants to remove soybean lecithin and phenol in an insulin-loaded emulsion system (IES). The results showed that n-pentanol was a suited extractant to meet the measurement requests because a large quantity of soybean lecithin and phenol were removed simultaneously, and there appeared no interferences brought by n-pentanol in insulin measurement. The analytical method was assessed by the criteria including linearity (linearity range: 1.05-7.34U/mL and correlation coefficient: 0.99936), accuracy (recoveries: 98.1-102.5%), within-day precision (R.S.D.: 0.5%, n=6), LOD (0.01U/mL), and LOQ (0.03U/mL), and exhibited high sensitivity and specificity, and considerable accuracy and precision on the determination of insulin in the emulsion system. The application of this method had been successfully performed to study the storage conditions of IES, and the results indicated that insulin in IES would be stable at 4^oC in light proof condition, which was significant for further pharmaceutical research.

            Description:
Perchlorate can be determined by the tandem technique of ion chromatography (IC) coupled to electrospray ionization mass spectrometry (ESI-MS). However, detection by ESI-MS can be compromised by the coelution of matrix components that can suppress the analyte signal. In addition, the presence of surface-active and other types of matrix components can cause fouling of the electrospray inlet, reducing overall signal and requiring frequent maintenance. The influences of matrix components can be minimized by using analytical columns with different selectivities, in-line diversion of separated matrix components, and off-line selective removal of matrix components via ion exchange or adsorption. This paper will discuss these sample preparation approaches for samples containing anionic species including surfactants and inorganic ions that elute in the vicinity of perchlorate.

              Description:
A simple and cost-effective capillary electrophoresis/mass spectrometric (CE/MS) method for the analysis of small carboxylic acids including succinate, malate, tartarate, maleinate and citrate, is described. All CE/MS experiments were performed with uncoated fused-silica capillaries and with alkaline volatile buffer solution (ammonium formate buffer, pH 10). Since sheath liquids have significant effects on the sensitivity in typical CE/MS applications, the effects of type and flow rate of the sheath liquids on the sensitivity of carboxylic acids were investigated. As the result, the best sensitivity was obtained with the alkaline sheath liquid (5mM ammonium hydroxide in water/methanol (50/50, v/v) solution) at 6@mlmin^-^1. With the alkaline volatile buffer solution, sufficient electroosmotic flow (EOF) to carry all small carboxylic acids toward the cathode (MS side) was obtained, although all analytes had different electrophoretic mobilities toward the anode (the CE inlet). Taking advantage of the relatively higher EOF velocity, several carboxylic acids could be detected by MS in ESI-negative mode with a short analysis time. The R.S.D. values (n=5) for the migration time and the peak area of the carboxylic acids tested were less than 0.6 and 4.2%, respectively. The method was applied to the CE/MS analysis of carboxylic acids in apple juice to demonstrate the applicability to real samples.

                Description:
Cylindrospermopsin (CYN) was determined by liquid chromatography/electrospray ionization-mass spectrometry (LC/ESI-MS) using 2-[4-(2-hydroxyethyl)-1-piperazinyl]ethanesulfonic acid (HEPES) as the internal standard. In the selected ion monitoring of LC/ESI-MS, m/z 414 for CYN and 237 for HEPES were monitored using the negative mode; the retention times of CYN and HEPES were 12.41 and 14.21min, respectively. CYN was determined from peak area ratios of m/z 414/237. By the treatment of an anion exchange cartridge using a buffer at pH 10.5, CYN was isolated and condensed. No interfering peak was observed. Linearity of this method was observed at the range of 0.10-31.12ng. Total coefficients of variation were 5.1 and 2.9% at 104 and 1038@mgCYNL^-^1. The quantitative limit at a signal-to-noise (S/N) ratio of 10 was 0.16ng. CYN concentration in natural waters is low. CYN in waters should be condensed for determination. This method including the treatment for isolation and condensation of CYN is useful for determination of CYN in environmental and/or drinking waters.

                  Description:
Isotope dilution liquid chromatography coupled with electrospray ionization tandem mass spectrometry (LC-MS/MS) was successfully applied to the quantification of acrylamide (2-propene-amide) in infant rice cereals (with nutrients) and other cereal-based foods (fastfood nutrient cake, teething cookie, hi-protein milk wheat and vegetable teething rusk). The method included defatting with petroleum ether, extraction with 2M aqueous solution of sodium chloride, further liquid-liquid extraction with ethyl acetate and clean-up by solid-phase extraction (SPE) with OASIS HLB 6cc cartridges. Acrylamide was detected with an Atlantis dC"1"8 5@mm; 210mmx1.5mm column using 10% methanol/0.1% formic acid in water as mobile phase. The analytical method in the present study was well validated and good results were obtained with respect to repeatability (R.S.D. <6.5%) and recovery (87-96%) which fulfilled the requirements defined by European Union (EU) legislation. The acrylamide levels in infant rice cereals and other cereal-based foods were 3.3-37.1@mgkg^-^1 and 10.9-1568.9@mgkg^-^1, respectively. This validated method might need to be modified under some special conditions. For instance, an additional deproteinating step should be operated during the sample treatment process as for the analysis of some protein-rich infant cereals.

Hardbound. The seven reviews articles presented in this volume cover a broad range of subjects. The first article is concerned with the use of active optics in modern, large telescopes. The second article discusses variational methods used in nonlinear fiber optics and in related fields. The article by O. Keller which follows deals with a topic of historical interest. The article presents a fascinating account of researches of the Danish physicist L.V. Lorenz who in 1867 established the electrodynamic theory of light, independently of the work of James Clerk Maxwell. The fourth article is concerned with the canonical quantum description of light propagation in dielectric media. The fifth article by D. Dragoman describes the similarities and the differences between classical optics and quantum mechanics in phase space. The article by R. Boyd and D. Gauthier which follows, summarizes recent research on pulse propagation effects in resonant material system. T

                                  Book Description

The residents of Toehold, Alaska, are an odd collection of eccentric souls reveling in the fierceness of the land and determined to live on their own terms. All of them have stories, some ridiculous, some bordering on the Homeric. Summer Joe has a harem of wives and a weakness for Jewish social workers. Six-foot-three-inch Sweet-ass Sue runs the town's only bar. There's Buddy Barconi, an ex-New York fireman with a raunchy sense of humor matched perfectly by a total lack of propriety. And Mary Ellen Madden, known as Mel to her friends, a cash-strapped vagabond with gray-green eyes and a double-wide smile who's trying her luck as a hunting guide.

When her first customer appears, a ruthless Hollywood producer seeking the glory of the kill, Mel and her best friend, Cody Rosewater -- the son of a San Francisco flower child, and now Toehold's taxidermist -- can no longer ignore the tension that's been crackling between them for years.

Always in the background is the raw majesty of the northern wild: crazed moose bolt down Main Street, caribou are violently ambushed by wolves, and the eight-month-long winter night is illuminated by the northern lights.

Funny and romantic, with a cast of unforgettable characters, Toehold is at once a laugh-out-loud comedy, a quirky love story, and a sublime evocation of the beautiful, rugged wilderness of Alaska.

                                  Book Description

Alan Johnson is a man ill-at-ease among people and only slightly more comfortable with animals. His story begins in upstate New York with his rescue of an injured Dalmatian who "came down out of the sky and survived the fall, showed me how gradually I have fallen--how I never touch, never really talk to another person...I am hardly a person at all." The dog heals and is returned to its neglectful owner, but Alan Johnson steals it back and heads west in search of what it means to be human.

As he crosses the United States, he moves through landscapes full of animals half-tamed and people run wild: a fanatical taxidermist, a lonely woman raising tigers on her remote ranch, a tragic circus chimp named Rufus, contemporary polygamists, and the caretakers of boot camps for troubled youths. They are Carnival Wolves, manifestations of our attempts to tame what is dangerous and wild, distorted reflections of parts of ourselves.

After a tortuous journey through various states of depravity--and of America--Alan Johnson ends up in California having reached a reconciliation of instincts and having found a human being he can love. A gripping, hallucinatory read, Carnival Wolves is a provocation, a plea for identification that questions the humanity of its readers and confirms Peter Rock as a unique literary talent.

Carnival Wolves is an average collection of tales that could have been a first-rate novel or selection of short stories. Rock tries to be too artsy for his own good at times. He has a very interesting story to tell and some compelling symbols to convey if he just wouldn't get in his own way! Rock will be a great novelist if he just lets himself...

5 out of 5 stars Peter Rock is the real deal.......1999-12-06

This book is magic, its language like incantation--proof of a brilliant mind and hands at work. I have read it a number of times and its organization is uncanny and ingenious, how it can operate like a novel and a short story collection. The chapter entitled Death's Door is one of the most finely crafted stories I've ever read anywhere. Another chapter was lauded by the O. Henry Award folks. Do yourself a favor. Read his book and tell a friend. Mr. Rock is deserving of the recognition.

This book takes you on cross-country tour of America, shows you some believable and interesting people, in places on and off the beaten track. The people described are the mainstream america we have been waiting to be shown. Each chapter is a story that can stand on its own, which makes for great summer reading.
