                Book Description

Arabidopsis has long been acknowledged as the 'Botanical Drosophila' with its small genome, low levels of repetitive DNA, small size and fast generation time it is an ideal molecular genetic tool for the analysis of development in higher plants. Arabidopsis: A Practical Approach provides an introduction to most of the key techniques required for the use of Arabidopsis as an experimental system. It gives a basic introduction to the optimal growth conditions and genetic resources available for Arabidopsis, how this material should be handled, maintained and used. Individual chapters describe strategies for the identification, mapping (using multi-marker lines and recombinant inbreds), and characterisation of different mutants by microscopy, molecular cytogenetics and gene expression analysis. Different cloning strategies, using transposons, T-DNA and map position are described in detail. Sequencing of the Arabidopsis genome will be completed in 2000 and bioinformatics are of key importance; the tools that are available and where they can be found on the Web are presented.

                  Book Description

Loose-leaf Binder. 'Cloning Vectors', the very first comprehensive compilation of the presently existing vectors used in cloning, was published in December 1985. As a laboratory manual, it serves in rapidly identifying a given vector best suited for a particular purpose. Because of the extremely rapid developments in the field, annual supplements are published (starting from 1986), which comprise newly identified vectors. The first two supplementary updates are now available.

A large section of the manual is devoted to Escherichia coli, the most commonly used host for general cloning and manipulation of DNA sequences. However, other Gram-negative bacteria are also discussed, as are several Gram-positives, such as Bacillus subtilis, the genera Staphylococcus, Streptococcus and Streptomyces, the lower eukaryotic yeasts and fungi, and animal and plant cells in culture.

Within each chapter on these host organisms, separate sections have been created for general a

                    Book Description

First published in 1988 and now known as a classic. In this book Stephen Fry describes, in bench-top detail, techniques for the study of primary cell walls in higher plants. Deepening interest in the chelistry and metabolism of cell walls reflects the breadth of their significance in biology. Walls play crutial roles in morphogenesis, growth, disease-resistance, recognition, signalling, digestibility, nutrition and decay. Scientists interested in questions concerning these processes need to know about methods of cell wall analysis. This book was written with these people in mind. There are 11 chapters as follows: 1. Introduction to the growing plant cell wall 2. Radioactive labelling of cell walls 3. Wall polymers: extraction and fractionation 4. Wall polymers: chemical characterisation 5. Wall biosyntesis 6. Wall enzymes 7. Wall architecture 8. Wall turnover and sloughing 9. Biologically-active wall oligosaccharides 10. Wall tightening and loosening: growth 11. Wall degradation "Stephen Fry has written an invaluable book on the analytical techniques available in this important research field. This is an excellent book for the typical modern biologist." J. Experimental Biology "This book will be of considerable value to anyone engaged in the analysis of plant cell walls." Ann. Botany "For those who intend to do any practical cell wall analysis this book will be an invaluable laboratory manual." Plant Pathology "As a practical guide to wall analysis this book will be a most valuable tool." Times Higher Education Supplement

It has been reproduced in the best form available to the Pentagon. It is not spiral-bound, but rather assembled with Velobinding in a soft, white linen cover. The Storming Media report number is A675023. The abstract provided by the Pentagon follows: Normal biologic function of an organism requires communication and regulation at several levels. Perhaps the most fundamental communication is that which occurs between the extracellular matrix and the intracellular environment. Elucidation of the signal transduction pathways in the areas of chondrocyte, osteoblast and inflammatory regulation would have significant implications in wound healing, regenerative procedures, implants and treatment of metabolic diseases of bone and cartilage. The rat costochondral chondrocyte model has been used to demonstrate that vitamin D3 metabolites can directly regulate many cellular and extracellular matrix functions and that this regulation is dependent both on the type of vitamin D metabolite, and on the maturation level of the chondrocytes. Matrix vesicles, distinct extracellular organelles critically involved in matrix mineralization, are also differentially regulated by vitamin D metabolites. Recently, it was shown that vitamin D metabolites can regulate protein kinase C activity in chondrocyte culture lysates and that these effects are also metabolite- and maturation- specific. Protein kinase C (PKC) is a family of isozymes that are known to be important in cell signal transduction. The aim of this study was to examine whether PKC mediates the nongenomic effect of vitamin D on chondrocyte regulation. The present study investigated whether PKC activity was present in matrix vesicles (MV) and plasma membranes (PM) and whether this activity can be directly regulated by vitamin D metabolites.

The Storming Media report number is A076604. The abstract provided by the Pentagon follows: This dissertation describes a series of toxicological studies carried out in order to better understand the role of monoamine-active drugs in drug- related deaths. A simple, robust method for determining psychiatric drugs and metabolites was devised using positive mode APESI LC-MS at basic pH. This method was modified to enable determination of multiple commonly abused drugs. It was discovered that both psychiatric drugs and drugs of abuse could be detected using similar chromatographic parameters. Target drug and metabolite concentrations were determined in postmortem tissue specimens. Drug concentrations were highest in bile and liver and lowest in vitreous humour. Four patterns of distribution were observed, with extent of biliary excretion corresponding to molecular weights. Concentrations in other tissues were compared to those in blood. Several significant correlations were observed, suggesting target drug concentrations in blood could be predicted using these tissues, or could be analysed in lieu of blood in cases of extensive exsanguination or putrefaction. Antipsychotic concentrations were determined in various brain regions from schizophrenics to determine whether such drugs partition preferentially into particular regions. A further aim was to investigate whether trends exist in how they distribute in brain as a drug class. It was found that if regional drug concentrations were normalised for those in cerebellum, they distributed into three distinct patterns, corresponding to structural features of different phenothiazines.

                                Description:
In this paper, the selectivity and sensitivity of cyclodextrin (CD) modified infrared (IR) chemical sensor in detection of aromatic acids in aqueous solutions were reported. To eliminate the interference from water, the technique of attenuated total reflection was employed. By surface treated with CD molecules on the internal reflection elements, the sensors were selective in sensing of aromatic acids compared to aromatic compounds with other functional groups. To facilitate the use of this method for the quantitative analyses of aromatic acids in aqueous solutions, analytical functions were also developed in this work and a linear relationship between analytical responses and concentrations of analytes can be obtained. To optimize the analytical conditions, the factors that influence the IR spectroscopic signals were examined. These factors included response time, CD loadings of the sensors, pH effect on response, regeneration efficiency and stability of sensors. Under the optimal conditions, the detection limits for aromatic acids at a detection time of 2min can be <100@mg/L. Meanwhile, the dynamic linear range for detection was only ca. two orders of magnitude if direct IR signals were used. Using the analytical function developed in this work, the linearity can be extended up to a concentration of 100mg/L.

                                  Description:
To explore the properties of cyclodextrins (CDs) as an optical sensing phase, the behavior of immobilized CD in interaction with analytes was studied in this work. CDs having different cavity sizes were immobilized onto the surface of infrared (IR) internal reflection-sensing element (IRE) to kinetically monitor the behavior of CD in interaction with analytes. Several aromatic compounds having various molecular sizes and functional groups were used to characterize the interaction mechanism. A two-layer modification method was proposed in this work, which utilized a thin hydrophobic film (polyvinyl benzyl chloride) to stick on the IRE and to covalently bond to the CDs through an ethylene diamine linker. The synthesized CD phases exhibited high stability in aqueous solution. To analyze the behavior during the formation of complexes between the guest molecules and the CD phases, we modeled the interaction behavior and treated the kinetic data with the theoretical equations developed in this work. The results indicate that the behavior of the interaction between guest molecules and CDs was explained by considering the formation of two types of complexes: adsorbed complexes and inclusion complexes. The formation of the inclusion complexes was relatively fast, the time required to reach equilibrium could be shorter than a few minutes. The adsorbed complexes were also observed, but their rate of formation was relatively slow; equilibrium could be reached at times greater than 60min. Based on the signals observed under equilibrium conditions, the concentration of inclusion complexes was approximately three times than that of the adsorbed complexes.

In describing this book to a friend I said, "Well, it's about a girl who has an affair with a much older man, and there's a sex scene involving a banana, and a talking cat who believes that he and the main character were lovers in their prior lives as fishmongers. Or something like that." Couldn't bear to finish, but my fellow bookclubbers tell me the ending was a terrible disappointment.

                                        2 out of 5 stars Elliot's Banana.......2005-12-30

The cat was totally wierd! It was okay for a quick read but if you are looking for something deep this isn't it. I think that the author tried her best to make it something it wasn't but come on we have all the read books where someone has lost a loved one and it takes them less time to get their life back in less than 200 pages.

5 out of 5 stars Three stories that came together to make one great novel.......2005-09-12

According to the Q & A by the author in this book, she originally started out writing three short stories and realized there was a way to intertwine them into this moving novel. After reading this book, I'd say she did a terrific job.

Junie knows that she's still trying to figure things out. Temp jobs by day and the girlfriend of the drummer in a local band. 25 and just moved in with Leon, the drummer. All of this fun and in NYC. Though truth be told we dont find this out about her until we meet Eliot.

Eliot, the sci-fi writer who has a cat with a story all of his own. Alfie, the cat who believes he's been reincarnated. And it is because of this cat that Eliot and Junie meet. Eliot, who is twice as old as Junie, intrigues her beyond anything she's ever known before. Junie is captivated by how quickly her life changes when she meets this writer.

The story progresses and Junie's character becomes more evident. Still burdened by the loss of her brother when she was 8, Junie believes that her whole life she's trying to find out who she is and what she's supposed to do. In the matter of one weekend, her whole perspective changes. It isn't until ending both of her relationships and attemtping to rekindle one that she thought was long gone did she realize who she was and what she was going to do.

Junie may not have things together but her transformation from taking comfort in the past to confronting her present brings a great deal of insight to this novel. The title to this book is quite appropos on many levels. From the innuendo that starts the book off and it is the understanding of how to avoid a slippery peel to confront those things in your life that trouble you.

This may not be your traditional 'chick lit' fare but it certainly is worth the read. Humor and wit, love and heartache, this novel has it all.

2 out of 5 stars Not deep at all.......2005-03-24

While there were some touching moments, for the most part this book was weak. I disliked how long it took the protagonist to find out "what was wrong with her". How about some introspection? It was very obvious. Also the voice of Eliot's cat throughout was highly annoying and distracting.

3 out of 5 stars How about alittle hot fudge with this banana?.......2004-12-30

I found myself wondering why Junie couldnt seem to get it right through most of this book. She seems to float along aimlessly in and out of Leons life into Eliots then back to Leon. The only thing that she seems to get any pleasure from is the banana or does she?
All in all this was a very strange book.
