Bridging the gap from physics to cell biology.......2002-02-20
I wish there was a way to give a book more than five stars, because if there was, this book would get them! One of the most important trends in cell biology in recent years is the study of mechanics at a single molecule level. Since most of the really interesting processes in cell biology, such as division and motility, ultimately are carried out by molecules that convert energy into motion, the question of how these molecules actually move has very broad implications and impinged on everyone. However, in order to understand this, one has to understand some basic physics which of course involves some math. The biologist who is interested in this subject thus has two, and only two options. one is to bury one's head in the sand and say its just too hard to understand, and the other is to read Joe Howard's book. Although the mathematical content is readily apparent, only the basics are needed to get started. The author has thoughtfully compiled detailed derivations at the end in an appendix, so that one can see the details without becoming enmired in them on first reading. Considering the potential difficulties of taking a "hard science" approach to cell biology, the book is remarkably easy to read, which is a tribute to the thought the author has put into presenting the subject in the most logical possible way.
In addition to being an excellent entry point for biologists into this subject, this book would also be an excellent resource for engineers who become interested in cell biology (like myself) because it presents many of the current research frontiers in cell biology from an essentially engineering perspective and using quantitative reasoning. Again, the author has taken great pains to present the subject in a logical way that does not require much prior knowledge about biology on the part of the reader. Thus, either for biologists who want to learn about the quantitative/physical approach to cell biology, and for engineers or physicists who want to learn how they can apply their type of thinking to problems of cell biology, this book is highly recommended.

This book presents new mathematics for the description of structure and dynamics in molecular and cellular biology. On an exponential scale it is possible to combine functions describing inner organisation, including finite periodicity, with functions for outside morphology into a complete definition of structure. This mathematics is particularly fruitful to apply at molecular and atomic distances. The structure descriptions can then be related to atomic and molecular forces and provide information on structural mechanisms. The calculations have been focussed on lipid membranes forming the surface layers of cell organelles. Calculated surfaces represent the mid-surface of the lipid bilayer. Membrane dynamics such as vesicle transport are described in this new language. Periodic membrane assemblies exhibit conformations based on the standing wave oscillations of the bilayer, considered to reflect the true dynamic nature of periodic membrane structures. As an illustration the structure of an endoplasmatic reticulum has been calculated. The transformation of such cell membrane assemblies into cubosomes seems to reflect a transition into vegetative states. The organisation of the lipid bilayer of nerve cells is analyzed, taking into account an earlier observed lipid bilayer phase transition associated with the depolarisation of the membrane. Evidence is given for a new structure of the alveolar surface, relating the mathematical surface defining the bilayer organisation to new experimental data. The surface layer is proposed to consist of a coherent phase, consisting of a lipid-protein bilayer curved according to a classical surface - the CLP surface. Without employing this new mathematics it would not be possible to give an analytical description of this structure and its deformation during the respiration cycle. In more general terms this mathematics is applied to the description of the structure and dynamic properties of motor proteins, cytoskeleton proteins, and RNA/DNA. On a macroscopic scale the motions of cilia, sperm and flagella are modelled.
This mathematical description of biological structure and dynamics, biomathematics, also provides significant new information in order to understand the mechanisms governing shape of living organisms.

A Classic.......2007-03-02
The Methods in Enzymolgy series is probably the most useful and respectable series of books for those of use who actually work in this field. This volume is a classic, even at almost 30 years old it is still excellent.
Book Description
The purpose of this book is to present a full spectrum of views on current approaches to modeling cell mechanics. In part, this diversity of opinions stems from the different backgrounds of those who have contributed to the field. The authors of this book come from the biophysics, bioengineering, and physical chemistry communities, and each joins the discussion with their own unique perspective on biological systems. Consequently, the approaches range from finite element methods as commonly used in continuum mechanics, to models of the cytoskeleton as a cross-linked polymer network, to models of glassy materials and gels. Studies reflect both the static, instantaneous nature of the structure as well as its dynamic nature due to polymerization and the full array of biological processes. It is unlikely that a single, unifying approach will evolve from this diversity, in part because of the complexity of the phenomena underlying the mechanical properties of the cell. It is our hope, however, that a better appreciation of the various perspectives will lead to a more highly coordinated approach to these essential problems, and might facilitate discussions among those with differing views.

Actin is an extremely abundant protein that comprises a dynamic polymeric network present in all eukaryotic cells, known as the actin cytoskeleton. The structure and function of the actin cytoskeleton, which is modulated by a plethora of actin-binding proteins, performs a diverse range of cellular roles. Well-documented functions for actin include: providing the molecular tracks for cytoplasmic streaming and organelle movements; formation of tethers that guide the cell plate to the division site during cytokinesis; creation of honeycomb-like arrays that enmesh and immobilize plastids in unique subcellular patterns; supporting the vesicle traffic and cytoplasmic organization essential for the directional secretory mechanism that underpins tip growth of certain cells; and coordinating the elaborate cytoplasmic responses to extra- and intracellular signals. The previous two decades have witnessed an immense accumulation of data relating to the cellular, biochemical, and molecular aspects of all these fundamental cellular processes. This prompted the editors to put together a diverse collection of topics, contributed by established international experts, related to the plant actin cytoskeleton. Because the actin cytoskeleton impinges on a multitude of processes critical for plant growth and development, as well as for responses to the environment, the book will be invaluable to any researcher, from the advanced undergraduate to the senior investigator, who is interested in these areas of plant cell biology.

The underlying hypothesis of this book is that the fibers are orientated by self-assembly just outside the cells during a mobile liquid crystalline phase prior to stabilization; the author demonstrates that the commonest orientations of the fibers are plywood laminates (orthogonal and helicoidal), and as parallel fibers. Finally, he shows that these may be imitated in the laboratory by liquid crystalline chemicals. Many fine photographs will aid the initiated in recognizing the various kinds of fibers.
