Book Description

This book describes the most outstanding theoretical hypotheses aiming at explaining the infinite proliferative potential of a cell population, what is now known under the term cell immortalization. It elucidates the phenomenon from an evolutionary point of view, and describes the cell systems used and the experiments performed at the cellular and molecular levels to determine the mechanisms responsible for this cell property. It also appraises how the mortal and immortal phenotypes reflect characteristics of the respective organism, and stresses how this goes beyond the proposal that its opposite, the mortal phenotype, is a mechanism to protect the organism against cancer.

To test hypotheses on the origins of p53 mutations in human tumors, novel strategies are needed for generating mutation spectra experimentally. To this end we developed an assay employing Hupki (Human p53 knock-in) mouse embryonic fibroblasts (HUFs). Here we examine p53 mutations induced by aristolochic acid I (AAI)), the carcinogen probably responsible for Chinese herbal nephropathy. Six immortalized cultures (cell lines) from 18 HUF primary cultures exposed at passage 1 for 48h to 50@mM AAI harbored p53 mutations in the human DNA binding domain sequence of the Hupki p53 tumor suppressor gene. The most frequently observed mutation was A to T transversion, corroborating our previous mutation study with AAI, and consistent with the presence of persistent AAI-adenine adducts found both in DNA of exposed patients and in DNA of AAI-exposed HUF cells. One of the mutations was identical in position (codon 139) and base change (A to T on the non-transcribed strand) to the single p53 mutation that has thus far been characterized in a urothelial tumor of a nephropathy patient with documented AAI exposure. Of the seven p53 mutations identified thus far in >60 HUF cell lines that immortalized spontaneously (no carcinogen treatment), none were A:T to T:A transversions. In addition, no A to T substitutions were identified among the previously reported set of 18 mutations in HUF cell lines derived from B(a)P treatment in which transversions at G:C base pairs predominated.

A mixed bag.......2005-09-23

Book was in good condition, more or less as reflected in the price. However, it took a week and a half to arrive. My previous mail experience with Amazon book sellers has been much better -- none have taken more than 5 days for delivery.

Unimpressive.......2004-09-15

The level of detail and generality would make this book suitable for a lower-level undergrad course on urban planning or urban geography. However, even as someone who has completed an advanced degree, I found this book to be extremely difficult to follow. The topics covered are good; the photographs are well done. But the text itself is hard to understand. The logic of the author's argumentation is faulty in some places. Several diagrams left me clueless because there was not enough supporting documentation to interpret them. On the bright side, I did like his suggested portfolio assignments at the end of every chapter. If you can find a better book for your course, use it. Otherwise, this one will have to do.

Post Modern Mania.......2003-03-07

I thought this book gave a wonderful account of the development of American cities in a text book format. I enjoyed Knox's post modern slant on the process of urbanization.

Able to "see" spacetime.......2004-01-10

This is a beautiful, poetic, and generous book. I found that after reading and mulling over each section, I could eventually visualize how space was behaving under the influence of mass, and could also see why one needed to choose one's point of view carefully to understand the physics.

The book is intended as an introduction to general relativity, without much tensor formalism, but it does not neglect the geometrical underpinnings. It presents a very nice way to get at the physics, with creative and light-hearted diagrams.

I'm not so sure that Wheeler's own poems, which introduce many of the chapters, are a great success, but I admire and salute him for including them. Even his poetry helps to open additional doors to understanding general relativity.

A book that is truly beautiful.

Awesome: from an MSEE.......2002-08-25

The interrelation of gravity and spacetime is a formidable subject to describe; the author does so with excellence. The diagrams and charts reinforce the understanding.

Unfortunately when a key subject left me rather clueless, (Boundary of a Boundary) I spent quite a few frustrated hours being uncertain on whether or not to continue reading without the support of the material on those pages. As it turned out, the subject became clearer once I read on and returned again. I never did grasp it as completely as the rest of the book.

The book contains the most enlightening description of transverse wave propagation I've ever seen. It also helps solidify one's understanding of interval and relativity.

Not a book to be read overnight.

The author's mind has no boundary...........2002-02-04

This author is one of the most briliant, the most optimistic, and the most enthusiastic writer in all of physics, and in this book, his competence as a physicist and his deep fascination with the physical world is brought out dramatically. He is clearly a man who is feeling a powerful sense of exhiliration of the discoveries now taking place in all areas of knowledge. His foundation and his theme in the book is a simple geometric principle, namely that the boundary of a boundary is zero. He then guides the reader, assumed to have a rudimentary knowledge of mathematics, in a splendid presentation of the power of this principle in gravitational physics.

The first chapter is an overview of the history behind the subject, via the work of people who contributed to our current understanding of gravity. And then, with a masterfull use of diagrams he gives the reader a taste of the simplicity of the equivalence principle and the need to tack on an additional dimension (time) to the 3-dimensional space of everyday experience. The Pound-Rebka experiment is discussed as one that illustrates the idea of the spacetime interval, and the role of time dilation is discussed via the possibility of practical space travel. And such enthusiasm in his dialog: "the universe will grow ever more exciting", he says, and looking at the developments now taking place in today's science, he is indeed correct.

Chapter 4 gives a fascinating overview of what the author calls the boomerang, which illustrates the action of curvature on nearby test masses. This thought experiment involves the motion of a spacecraft through an imaginary tunnel through the Earth. The author analyzes the motion from the standpoint of Newtonian physics and general relativity. Curvature as the "grammar of gravity" is the topic of the next chapter, with illustrations of the paths of ants on spaces of zero, positive, and negative curvature. A very intuitive treatment of parallel transport around a closed path on a curved surface is given. The tides are discussed as a natural manifestation of the gravitational influence of the Moon on Earth.

Must difficult for a layman to understand is how spacetime acts on masive objects, but the author explains it brilliantly in the next chapter, taught via the concept of "momenergy". This entity is a 4-vector, and the author uses it to show how its creation in a spacetime region can be written as the sum of 8 terms, reflecting the fact that the "boundary" of a four-dimensional block in spacetime consists of eight three-dimensional cubes. That the contents of these cubes sum to zero is the famous "boundary of a boundary is zero", which is discussed in the next chapter. This chapter is one of the best explanations ever given (at this level) of the physics behind spacetime curvature and massive objects. The actual mathematical quantification of curvature is detailed in chapters 8 and 9, using elementary mathematics. The author discusses nicely the famous Scharwzschild geometry.

Concepts of a more concrete nature are discussed in chapter 10, wherein the author discusses the famous Pound-Rebka experiment and planetary motion. This is followed by a discussion of the elusive gravitational waves in chapter 11. Again with a clever use of illustrations, the author explains the transverse property of gravitational waves, and compares gravitational waves with electromagnetic waves. The role of the quadrupole moment in the creation of gravitational waves is brought out briliantly by the author. He discusses briefly various attempts to detect gravitational waves.

Black holes are the topic of chapter 12, wherein the famous Penrose process for extracting energy from a black hole is discussed, and the "no-hair" theorem for black holes. A neat symbolic representation of the Bekenstein number of a black hole is given. The role of the Hawking process, connection quantum processes with the physics of black holes is briefly discussed. The author ends the book with a look at the expansion of the universe, the missing mass problem, and another very interesting topic that has gained much attention recently: the concept of gravitomagnetism. This is a "weak-field" prediction of general relativity, and predicts that the rotation of the Earth should influence the motion of orbiting satellites. This topic is currently bringing together ideas such as the quantum Zeno effect, Mach's principle, and the notorious "frame dragging" effect in general relativity. Experiments do measure it are currently in play and in the proposal stage, namely the LAGEOS and LAGEOS II experiments, which measure the gravitomagnetic orbital perturbation, which is known as the Lense-Thirring effect.

A stunner of a book.......1999-12-13

This is truely an amazing book. Wheeler does for General Relativity what Hawking did for Cosmology in "A Brief History of Time", and in some sense they are similar books. However Wheeler has a unique, quirky style of writing that makes the book an entertaining adventure to read.

Wheeler is able to pull off a major accomplishment: He explains Einstein's General Relativity in a clear, straightforward manner, with a minimum of math. It's "conventional wisdom" that General Relativity is seriously serious stuff, the domain of hardcore Physics geeks. That doesn't faze Wheeler. He leads the reader along, gleefully pointing out the scenery, making it all look quite simple and understandable. And then all of a sudden, when you least expect it, you find he's derived and presented Einstein's field equations with only a teensy-tiny bit of algebra! Even if you know this stuff already, his presentation makes you think about it with a new perspective.

And did I mention the illustrations? They are really exceptional.

If you have any interest or dealings with GR, ya gotta have this book!

A pleasing mixture of gothic and nautical horror.......2004-11-11

At the beginning of the twentieth century, F. Marion Crawford was one of the most prolific, widely-read novelists of the English-speaking world--a sort of Sidney Sheldon of the Edwardian era. Now, his novels are banished to the musty shelves of old-fashioned romance and historical fiction, but his supernatural tales live on, most especially "The Upper Berth" and "The Screaming Skull."

Lee Weinstein has collected all eight of Crawford's supernatural stories, some of them gothic in the extreme, others oozing pathos. But this author was at his best when writing of the sea and its unforgiving dead. Many of his stories retain a place of honor in ghostly anthologies for their atmosphere of slowly-building horror. I am in agreement with Lee Weinstein when he says, "One can only regret that he did not write more of them."

"The Dead Smile"--A gothic tale of forbidden love and vengeance from beyond the tomb. We 21st century readers are a bit more used to dealing with the theme of incest, but when Crawford published this tale, it must have shocked many Victorian sensibilities. Incestuous hints abound. The evil, dying Sir Hugh Ockram, his son, and his son's fiancée all have the same hellish smile: "...She smiled--and the smile was like the shadow of death and the seal of damnation upon her pure, young face." The best scenes are in the vault below the castle, where the Lords of Ockram lie in burial shrouds, but not entombed.

"The Screaming Skull"--A doctor murders his wife by pouring molten lead in her ear. He dies mysteriously with his throat torn out. The old seaman who inherits the doctor's cottage also inherits a skull in a hat box. Something inside the skull rattles when he shakes it. When he tries to get rid of the skull, the screams begin.

"Man Overboard!"--This story was worth the price of the book for me, because I'd never seen it before, and it's a great ghost story. Just let the obscure nautical language flow past you, e.g. "I coiled down the mizzen-topsail downhaul myself, and was going aft to see how she headed up..." This is a story of identical twin brothers who both love the same woman. When they go to sea on the 'Boston Belle,' one brother is swept overboard during a storm and drowns--but somehow remains part of the crew. As is true in most supernatural stories featuring a wedding, the innocent bride meets a horrible fate.

"For the Blood is the Life"--I don't much care for vampire stories, but this one is wonderfully eerie. Two men are dining on the roof of an old tower-fortress on the Southern Italian coast. After moonrise, the guest sees a figure lying on a mound of earth near the tower and goes to investigate. When he returns, his host tells him the story of the grave-mound.

"The Upper Berth"--A business traveler who makes many Atlantic crossings secures a berth on the 'Kamchatka,' bound for Liverpool. He requests a room with a double bunk and is disappointed to learn that he will have a roommate in the upper berth. The first night of the voyage, his roommate runs screaming out of the small room and throws himself overboard. The business traveler learns that three other men who booked into room 105 have killed themselves in the same fashion, and he is determined to investigate.

"By the Waters of Paradise"--A gothic tale that has a happy ending for a change. A melancholy young man is raised by his superstitious Welsh nurse in an ancestral castle, surrounded by gardens and fountains. One night the old nurse sees "One--two leaden coffins, fallen from the ceiling!" Sure enough, his parents die, and the nurse tells her charge the story of the Woman of the Water. Will the new lord of Cairngorm be able to escape the curse?

"The Doll's Ghost"--An old man repairs a rich girl's doll and becomes so fond of it he can hardly bear to part with it. Finally, he instructs his young daughter to return the doll to its owner. The daughter doesn't return, but the doll does.

"The King's Messenger"--A man is seated between a lovely young girl and her beloved at a dinner party, and learns that his right-hand neighbor is the King's Messenger. After the girl disappears, he discovers what the man really does.
