Book Description
The Ras superfamily (>150 human members) encompasses Ras GTPases involved in cell proliferation, Rho GTPases involved in regulating the cytoskeleton, Rab GTPases involved in membrane targeting/fusion and a group of GTPases including Sar1, Arf, Arl and dynamin involved in vesicle budding/fission. These GTPases act as molecular switches and their activities are controlled by a large number of regulatory molecules that affect either GTP loading (guanine nucleotide exchange factors or GEFs) or GTP hydrolysis (GTPase activating proteins or GAPs). In their active state, they interact with a continually increasing, functionally complex array of downstream effectors.
Since the last Methods in Enzymology volume on this topic in 2000, Rho GTPases have continued to receive a huge amount of attention. The human genome sequence has revealed the full extent of the Rho GEF and Rho GAP families (over 80 members for each) and the challenge of identifying the molecular interactions and cellular pathways influenced by each of these regulators is a daunting prospect. This new volume describes some of the methods currently being used to examine Rho family GTPase regulation at the biochemical and cellular level.

Work on signal transduction pathways involving Rho family GTPases started some ten years ago. There is now a realization that these highly conserved molecular switches not only serve to coordinate cytoskeletal organization but also impact on diverse aspects of cell biology such as membrane trafficking, transcriptional regulation and mitotic progression.
Humans contain more than 20 Rho type GTPases. This volume not only presents a detailed phylogenetic analysis of Rho proteins, but also discusses the possible origins of the human members. Such an analysis of human Rho GTPases has not previously been attempted.
The book includes an overview of how Rho GTPases become activated which is complemented by an extensive Chapter by Darerca Owen and Helen Mott who unravel the beautiful molecular details given to us by the many structural studies of Rho GTPases.
The key areas currently being investigated in relation to these ubiquitous proteins are described for both in vitro and in vivo systems. These are presented in a format that ensures the reader can approach the topic with minimal background knowledge, while ultimately bringing the subject to the level of an expert. Timely and highly authoritative, this volume illuminates newer findings, particularly as they relate to Rho proteins in vertebrate biology.

Weakly Nonlocal Solitary Waves and Beyond-All-Orders Asymptotics: Generalized Solitons and Hyperasymptotic Perturbation Theory represents the first thorough examination of weakly nonlocal solitary waves, which are just as important in applications as their classical counterparts. The book describes a class of waves which radiate away from the core of the disturbance but are nevertheless very long-lived nonlinear disturbances. Specific examples are provided in the areas of water waves, particle physics, meteorology, oceanography, fiber optics pulses and dynamical systems theory. For many species of nonlocal solitary waves the radiation is exponentially small in 1/epsilon where epsilon is a perturbation parameter, thus lying `beyond-all-orders'. A second theme is the description of hyperasymptotic perturbation theory and other extensions of standard perturbation methods. These methods have been developed for the computation of exponentially small corrections to asymptotic series. A third theme involves the use of Chebyshev and Fourier numerical methods to compute solitary waves. Special emphasis is given to steadily-translating coherent structures, a difficult numerical problem even today. A fourth theme is the description of a large number of non-soliton problems in quantum physics, hydrodynamics, instability theory and others where `beyond-all-order' corrections arise and where the perturbative and numerical methods described earlier are essential. Later chapters provide a thorough examination of matched asymptotic expansions in the complex plane, the small denominator problem in Poincaré-Linstead (`Stokes') expansions, multiple scale expansions in powers of the hyperbolic secant and tangent functions and hyperasymptotic perturbation theory. This book will be of special interest to applied mathematicians, fluid dynamicists in mechanical and aeronautical engineering, electrical engineers interested in fiber optics, quantum chemists and atomic and particle physicists.

Strange, Beautiful Smoke.......2001-04-09
If my memory serves me well, I first learned of "Smoke and Other Early Stories" after reading an article about Douglas Messerli, founder of Sun & Moon Press in Los Angeles. Messerli spoke of his giddy discovery that the copyright on many of Barnes' short stories had run so he had the legal right to publish them. Though Barnes apparently was not pleased by this, this collection is the result. And God bless the result. These fourteen tales (puntuated by Barne's own strange and evocative pen-and-ink illustrations) should be read by anyone who loves well-crafted, provative short fiction; and it should be a must for those who are beginning writers. The first sentence of each story introduces you to a world where everyday people and things transmute inexplicably into something weird and dreamlike: "Every Saturday, just as soon as she had slipped her manila pay envelope down her neck, had done up her handkerchiefs and watered the geraniums, Paprika Johnson climbed onto the fire-escape and reached across the strings of her pawnshop banjo." (From "Paprika Johnson.") Sometimes, she sets the stage simply as with the first line of "What Do You See, Madam?": "Mamie Saloam was a dancer." As Messerli notes in the introduction, Barnes' stories were published in newspapers at a time (the first two decades of the 1900s) when the public expected to see short fiction in such venues. Reading this collection can only make you long for such an era.
