Chemokines and Viral Infection (Current Topics in Microbiology and Immunology)

Chemokines represent a family of over 40 small proteins that, for the most part, are secreted into the environment and function by binding to G protein-coupled receptors (GPCRs) that are expressed on numerous different cell types. When initially identified close to 30 years ago, these molecules were associated with various human inflammatory diseases and it was recognized that expression may be integral in leukocyte recruitment to inflamed tissue. Within a relatively short period of time, early participants within the field determined that these proteins displayed distinct and conserved structural features and exerted potent chemotactic effects on defined lymphocyte subsets. There are now four sub-families of chemokines identified based on defined structural criteria relating to the positional location of conserved cysteine residues within the amino-terminus of the protein. Chemokines are now recognized as important in numerous biological processes ranging from maintaining the organizational integrity of secondary lymphoid tissue to participating in various aspects of both innate and adaptive immune responses following microbial infection. The host response to viral infection represents a well-orchestrated ballet consisting of numerous participants with diverse roles in defense but with the ultimate goal of generating virus-specific lymphocytes whose job is to control and eliminate the invading viral pathogen from infected tissues. Over the years, an emerging picture has developed that indicates that chemokines and their receptors are intimately involved in development of effective host responses to viral pathogens. Chemokine expression is now associated with all facets of defense against viral infection including linking innate and adaptive immune responses.

This book deals with the central role of cytokines in the generalized inflammatory response of the host as the consequence of severe infection/endotoxin action. International specialists cover several aspects in 20 chapters starting with the agents responsible (endotoxin, superantigens) and recognition during cytokine induction. Further chapters deal with the signal transduction cascade, its modulation due to sex or genetic polymorphism, and the possibilities and problems in detection (including surrogate markers). Major targets of actions are covered in the chapters on coagulation/fibrinolysis, adherence molecules, vasoactive factors, apoptosis and metabolism. As not all actions of cytokines are beneficial, several chapters deal with the prevention of induction, modulation of the cytokine generation or scavenging cytokines including gene therapy approaches. Models are necessary for obtaining pathophysiological information and for testing therapeutic approaches, and thus all chapters deal with exper imental mo dels as well as clinical trials. The reasons why these have failed so far are the subject of the final chapter.

This exciting new text describes how cells normally regulate immunological and inflammatory reactions, and how the immune system is intimately related to other bodily functions. The authors consider the effects of lymphokines on non-inflammatory cells and tissues, including connective tissue and the neuroendocrine system, and describe the effects of neuroendocrine and peptide growth factors produced by non-inflammatory cells and tissues on the functions of immune cells. To highlight the factors regulating immunophysiological functions, they discuss the inflammatory consequences of endotoxin, immune complexes, and complement; the interactions of immunomodulating epidermal factors and immune tissue; the modulation interactions of immunomodulating epidermal factors and immune tissue; and the modulation of immunity by cytokines. The mechanisms by which the immune system normally contends with bacterial, viral or tumor challenges are examined, with an emphasis on basic concepts and key experimental results; and the cells directly involved in host-defense processes are discussed. Wherever possible, information about in vivo and in vitro human immune responses is presented.

This book's pages are so thin they rip at almost every turn of the page. The publisher took a stab at trying to keep college students from reselling their books by making this one rip to shreds as they use it. This item has to be put in a three ring binder. Dont buy this version get the bound version.

This book was required for one of my college classes. It provides the same information that is taught in high school but in a much better format. I liked this book so much I have kept it for my personal library (and no I'm not some math/physics freak).
