          Leach explains how to do serious molecular modelling. Naturally, this has to be done by computer simulations. The text is a blend of the physical principles and equations needed, and how to implement as code. For example, there is the Lennard-Jones potential for intermolecular attraction. But modelling using this often also involves a cutoff. The problem with the latter is that it can create discontinuities in both the potential and the force [which is the derivate of the former]. You can shown how to suitably modify matters to avoid these complications.

          Another familiar potential is the Morse potential, used to model bond stretching. Empirical but very useful, and in general quite adequate for most simulations.

          The text even has an example of modelling a high temperature superconductor. Partly to enhance the relevance of the narrative for practical applications.

          In this book, Andrew Leach has done a great job in putting in almost every important concept, sundry as well as significant, from the field of computational chemistry and molecular modeling. From basic but very useful concepts like atom types, Z matrices, and force field parametrization, to advanced topics like Ewald Sums and Low Mode Monte Carlo conformational searching, Leach gives due importance to everything. The discussions on quantum mechanics in the first few chapters are moderate on the mathematics without shying away from it, and provide just the right amount of detail. Later chapters cover the whole gamut of computational techniques, from Molecular Dynamics and Molecular Mechanics, to Moelcular Similarity and QSAR. Examples that are relevant in chemistry and biology are scattered throughout the book and illustrate every key idea. There are many good books for Computational Chemistry and Molecular Modeling, and some are good for a few topics, others for other ones. However, if one wants to get a grip on ALL important topics in the area, I think this is the most comprehensive reference that one can look up.

          Good:
          This book gives an excellent overview of molecular simulation techniques starting with quantum mechanics ab initio type calculations and going up through molecular dynamics and polymer simulation. It gives a good step up from your standard physical chemistry text (such as Atkins or Chang) to being able to read the literature on modern molecular modelling techniques.

          Bad:
          The treatment of many methods is somewhat superficial.
          The book was first written in 1996 and updated in 2000 - it is starting to get a bit out of date.

          Overall I would recommend this as a solid introduction and reference.

          If you could choose only one book about molecular modelling - this should be it. Everything is covered, more or less thourough, from ab initio to molecular docking, algorithms, force fields, molecular dynamics, etc. It is well written, but also works well if you want to look up single key words. The book can be read by novices to molecular modelling as well as it is useful for experts. I can highly recommend it.

          I'd like to recommend this book from the perspective of someone who is new to the field. I have only an informal background in chemistry and biology and an undergraduate physics degree that's 20 years old. Mr. Leach works through a broad range of material, from numeric solutions to the quantum equations for a molecule to algorithms for searching through the conformational space. His descriptions have to be concise in order to fit the enormous volume of material he has to cover, yet I found that I had no trouble following along. He often takes a historical approach. I found this effective. I would find myself wandering down blind alleys when examining the early solutions, then read the later art and have a greater appreciation for the problem than if the ultimate answer were presented first. I have been able to use much of the book as a practical guide in my work.

          Frankly, I'm amazed that someone with a professional life can find the time to put together a book of this scope.

Hydrogen-bonding in assemblages of biological molecules examined from the structural point of view is a main focus of the present book. The concept of the hydrogen bond structure is developed, as is the role of cooperativity, in stabilizing particular structures and hydrogen-bonding dynamics. The information presented is based on the extraordinary increase in quantitative structural data that has become available from X-ray and neutron diffraction studies of crystal structures involving small and large biological molecules. Included are structural data of particular types of hydrogen bonds as well as data for different types of molecules, such as carbohydrates, purines and pyrimidines, nucleosides and nucleotides, ices and hydrates, cyclodextrins, nucleic acids and proteins.

                          Of all kinds of satire, there is none so entertaining and universally improving, as that which is introduced, as it were occasionally, in the course of an interesting story, which brings every incident home to life, and by representing familiar scenes in an uncommon and amusing point of view, invests them with all the graces of novelty, while nature is appealed to in every particular. The reader gratifies his curiosity in pursuing the adventures of a person in whose favour he is prepossessed; he espouses his cause, he sympathises with him in his distress, his indignation is heated against the authors of his calamity: the humane passions are inflamed; the contrast between dejected virtue and insulting vice appears with greater aggravation, and every impression having a double force on the imagination, the memory retains the circumstance, and the heart improves by the example.

                          This book is a great deal of fun to read. It is lively, witty and amusing, as well as strikingly modern in displaying the vicissitudes of fortune in the character Roderick. These vagaries of fortune, from penury, to wealth, from imprisonment, to landed gent are also reflected in the vagaries in the moods virtues (or lack thereof) in our title character, thus lending Roderick, for most of the book, a three-dimensional aspect and not simply another cardboard cutout for an 18th century picaresque.-But the book does have its faults, particularly as we draw to what we foresee will be the inevitable end. It's just too pat for many modern readers to swallow. Or at least it is for this one. The Oxford edition's notes, while helpful in places, especially with nautical turns of phrase, and for those with a scholarly interest in the location of certain streets in the London in Smollet's day etc tend to become rather annoying at times, almost to the point of insulting the well-read reader's intelligence. Many times I found myself saying, "As if I could not have figured that out on my own from the context!" The book, not surprisingly, is at its best when it is at its most autobiographical and descriptive, particularly the passages of Roderick's first sea voyage. One of my favourite passages that illustrates the lively vitality and humour of both the character and the work comes when Roderick, feared to be dying of typhoid fever, is visited by a priest to make a last confession:

                          ""Without doubt, you have been guilty of numberless transgressions, to which youth is subject, as swearing, drunkenness, whoredom, and adultery; tell me therefore, without reserve, the particulars of each, especially the last, that I may be acquainted with the true state of your conscience...."

                          Roderick, a thoroughgoing, Scottish Ant-Papist will have none of it and soon recovers.

                          I am reminded of Joseph Conrad's short story "Youth" which I recommend to all who enjoy this book. - But, in the end, Conrad's story is the philosophically deeper and more true-to-life narrative than this one.-Again, the ending, for this reader, was just too pat and soppy. I am not trying to be a "spoiler" here and ruin the reading of the book and imperiling this review, by telling you potential readers what it is. You don't need me for that. You will have figured it out about a hundred pages before the end. And, for the record, I believe that this misguided idea of not being able to include the reviewer's analysis of a book's ending seriously handicaps the reviewer as well as insults the reader's intelligence. ---But, I have to abide by the rules in order that this review be posted. So be it.

                          Anyway, a delightful 18th century romp, until the predictable winding down.

                          4 out of 5 stars Bad Boy Makes Good Reading.......2000-09-07

                          If you ever saw Sheridan's play "The Rivals," you might have heard this book mentioned. It's the book the daughter doesn't want her mother to know she is reading.

                          It pre-dates Jane Eyre, but it has that poor-lonely-orphan-wronged-by-the-world quality to it. Picture Jane Eyre as a young, red-headed Scotsman with a thin-skin, bad temper and active libido. Roderick's going to London reminded me of D'Artagnan arrival in Paris in "The Three Musketeers". His mere presence is often a cause of confict. There are insults, fights, brawls, battles, sea clashes, duels, and some more insults. Roderick is the world's punching bag and his own pride and scheming won't let him say, "I've had enough." All the while his hot blood is leading him into haylofts, bedrooms and yet more trouble.

                          Will this 18th-century punk ever wise up? Read for yourself. I suggest the Oxford World's Classics version. I didn't expect the footnotes, but they were a great boon. The language is archaic in places, so keep a dictionary handy.
