Book Description

A collection of state-of-the-art methods for epigenetic analysis, including recent breakthrough techniques that have great potential in the rapidly expanding field of non-Mendelian genetics. The authors provide techniques for the analysis of chromatin remodeling, such as histone acetylation and methylation. In addition, methods in newly developed and especially promising areas of epigenetics, such as telomere position effects, quantitative epigenetics, and ADP ribosylation are covered. There is also an updated analysis of techniques involving DNA methylation and its role in the modification, as well as the maintenance, of chromatin structure. Of special interest are potentially revolutionary techniques. These include methods for determining changes in native chromatin, methods of microarray analysis applied to epigenetics, and methylation-sensitive single-strand conformation techniques. The methods are suitable not only for studying fundamental biological processes, but also for investigating possible therapeutic interventions and such diseases as cancer.

5 out of 5 stars Very Up To Date Report on Current Research.......2006-03-17

It has been ten years since the first edition of this book. In that decade interest in telomeres has skyrocketed as a relation between telomeres and cancer has become more likely.

As is common with fields that are developing so rapidly, no 'standard' textbook has been written because it would become out of date so fast. Instead this book is a compendium of articles from researchers from around the world. Each of the articles has been peer reviewed and selected for this book by the editiors, each of whom is a pioneer in the field. The articles come from laboratories literally around the world, where ever the most important basic research is being conducted.

Cold Spring Harbor Laboratory Press is a very fast publisher. The basic material was finally collected for the book in July of 2005. By November of that year the book was in print. This means that the research being described is current to within half a year -- remarkably short timing for a published hardback book.

        Book Description

Molecular biologist Elizabeth Blackburn--one of Time magazine’s 100 “Most Influential People in the World” in 2007--made headlines in 2004 when she was dismissed from the President's Council on Bioethics after objecting to the council's call for a moratorium on stem cell research and protesting the suppression of relevant scientific evidence in its final report. But it is Blackburn's groundbreaking work on telomeric DNA, which launched the field of telomere research, that will have the more profound and long-lasting effect on science and society. In this compelling biography, Catherine Brady tells the story of Elizabeth Blackburn's life and work and the emergence of a new field of scientific research on the specialized ends of chromosomes and the enzyme, telomerase, that extends them.

In the early stages of telomere research, telomerase, heralded as a potential cure for cancer and diseases related to aging, attracted the voracious interest of biotech companies. The surrounding hype succeeded in confusing the role of telemorase in extending the life of a cell with a mechanism that might extend the lifespan of an entire organism. In Brady's hands, Blackburn's story reveals much about the tension between pure and applied science, the politicking that makes research science such a competitive field, and the resourceful opportunism that characterizes the best scientific thinking.

Brady describes the science accessibly and compellingly. She explores Blackburn's struggle to break down barriers in an elite, male-dominated profession, her role as a mentor to other women scientists (many of whom have made their mark in telomere research), and the collaborative nature of scientific work. This book gives us a vivid portrait of an exceptional woman and a new understanding of the combination of curiosity, imaginative speculation, and aesthetic delight that powers scientific discovery.

              Book Description

Telomeres and Telomerase Chairman: Sydney Brenner 1997 Telomeres are the protective genetic elements located at the ends of chromosomes and are essential for correct chromosomal structure and function. They are not fully replicated by the conventional DNA polymerase system because DNA synthesis occurs only in the 5??? to 3??? direction and requires an RNA primer for initiation. Consequently, cells require a special enzyme to maintain the telomeric ends of chromosomes during each round of replication. This enzyme, telomerase, is a ribonucleoprotein that extends chromosome ends by adding short stretches of nucleotide repeats using a portion of its integral RNA component as the template. Recently, much excitement has been generated by the suggestion that telomerase, or rather the absence of telomerase and the resultant loss of terminal DNA, is a cause of human ageing. The evidence for this is twofold: the telomeres of certain cells in culture shorten during their lifespan; and immortalization of cells is associated, at least in some cases, with the maintenance of telomeres and telomerase activity. The latter observation prompted the analysis of clinical samples from patients with cancer and the demonstration that, in contrast to normal somatic cells, malignant cells possess telomerase activity. This is a unique book. Not only does it contain the latest experimental results from an international group of experts, but it also includes critical examinations of the current evidence, and discussions that attempt to identify the central and underlying concepts of this rapidly expanding field.

                Book Description

This book provides an in depth review of our current knowledge on the role of telomeres and telomerase in ageing, disease and cancer. Telomere shortening results in telomere dysfunction, which represents one of the basic aspects of ageing contributing to the accumulation of DNA damage during ageing. The book describes cell intrinsic checkpoints (senescence and apoptosis) and environmental alterations that limit stem cell function during ageing and disease. The book focuses on experimental data in mouse models and cell lines and, in addition, provides and overview on telomere shortening in human diseases and ageing. The book also describes future directions in research on adult stem cell ageing, telomeres, and cancer. In addition, the book points to potential targets for molecular therapies aiming to improve regeneration and stem cell function during ageing or to impair cancer cell proliferation.

A special focus of the book is on adult stem cells. There is emerging evidence that adult stem cell ageing impairs organismal fitness and survival and contributes to cancer formation (cancer stem cells). The book summarizes basic mechanisms of adult stem cell ageing. Moreover, the authors describe consequences of telomere dysfunction on stem cell function involving cell intrinsic checkpoints as well as environmental alteration of the stem cell niche.

The above subjects appear to be of utmost importance to ageing researchers and physicians. Due to the advances in medicine, food supply and housing a growing percentage of humans reaches a long lifespan (>70 years). At this age molecular mechanisms of ageing limit organ maintenance, fitness and survival. A detailed understanding of the molecular mechanisms underlying the ageing process is essential to further improve quality of life and ‘health span’ in the ageing human populations.

In summary, this book provides an excellent basis for academic teachers, scientists, and students interested in the areas of stem cell ageing, telomeres & telomerase, regeneration, and cancer. In addition, the book is of interest for physicians since the development of molecular therapies targeting ageing stem cells represents a promising approach to improve regenerative reserve and organ function during ageing and could also serve to target cancer stem cells.

                Book Description

John A. Double and Michael J. Thompson have collected a critically important series of novel and essential techniques for studying telomeres and telomerase. These readily reproducible methods provide cutting-edge tools to identify, measure, and analyze telomeres, to determine telomerase expression at the RNA level, to determine telomerase activity, and to detect potential modifiers of this activity. The techniques for assaying telomerase activity range from standard radiological TRAP assays to nonradioactive methods, from non PCR-based methods to techniques using real-time PCR. Telomeres and Telomerase: Methods and Protocols provides the core array of productive techniques needed today to develop telomerase inhibitors or diagnostic/prognostic telomerase markers.

This book should be called Telomerase Book. Very detailed and helpful on telomerase assays but nothing satisfactory on telomere assays actually there is not one single protocol on actual telomere length measurement (only one method for detection one for distribution).

If you research the activity go for it, is a good book but if you want information on the length like me this is not a book for you.

A wonderful book. I loved it as a child, and going back to read it as an adult I realize that there are a lot of things that I missed. I remember my teacher reading it aloud in class--wonderful!

The movie, although very nice, has only a superficial resemblance to the book. For one thing, it moves the time a generation or so back. For another, in the book both parents are alive, rather than Caracticus Pott's being a widower; consequently, there is no romance.

I could very well wish that a new movie be made, NOT a musical and following the original plot.

Just like when I was a kid at my moms knee listening to her read this to me I wsa again transported to another time. This audio is great. Kids and adults alike will fall inlove with this audio book. Some what diffrent than the movie staring Dick Van Dyke. which is always a welcome suprise.

It is a charming and delightful story that you will love to read to your children. I read this book to my 6 year old. She loved it!! We both wish we could find more books about this wonderful family and car.

                            5 out of 5 stars Not the movie--even better!.......2005-09-04

I remember the 1968 movie based on this novel fondly, but had never picked up the novel itself until just the other day. I was surprised at how different the book is. Although Chitty-Chitty-Bang-Bang herself is very similar in both, the plot of the story is much different. I enjoyed this simple little story, with Fleming's humorous asides, very much. Those asides reminded me of the "Series of Unfortunate Events" books I have read and I would be interested in learning if Lemony Snicket drew from Fleming's story style for his own series. This is a fun, quick story that most kids will enjoy. I do, however, fear that the admittedly crude illustrations in my original edition would not be as popular with modern children.

                            5 out of 5 stars A Delightful Ride!.......2005-04-28

The story of Caractacus Pott, his family, and that wonderful magical car is one of the best children's stories you will ever read. Ian Fleming, the creator of James Bond, tells a fantastic story about Chitty Chitty Bang Bang, the magical flying car that can even turn into a boat! It's a delightful story, filled with crooks and gangsters, and you won't be sorry you bought this fantastic little book. Should be added to every child's bookshelf. Just delightful! The book is so much better than the movie, eliminating that silly Vulgaria story. If you want to read the story Ian Fleming intended, you'll have to buy the book. You won't be sorry.

When most people think of Ian Fleming, they think of "James Bond." This is the only children's book he ever wrote, and one wishes that he wrote more.

Forget the Dick VanDyke film, which has very little resemblance to the book. (Not that it is a bad film, mind you, taken by itself.)

In the film, Mr. Potts was a widower; in the book, Mrs. P. was very much alive. They pushed the time-frame of the book about 75 years, also, and (aside from using the idea of the rehabilitated racecar that could convert into a boat or an airplane) the plot had very little resemblance to that of the book.

The story is fairly simple, but is well told, rather in the manner of a parent telling a bedtime story. The author is not afraid to use big words, but he explains them. I remember at about age eight (shortly after I heard my teacher read it out loud to us in school) I used the word 'transmogrification' at home; my Mother told me that wasn't a real word, and I said it was, and we looked it up in the dictionary--and there it was! (Mother wasn't a stupid or uneducated woman; she was a college professor.)

The book could be read TO a child as young as four or five, and could be read BY a child of nine or up.
