Ao ler esse livro pude perceber que existe variadas técnicas que auxiliam na interpretação dos espectros de massas. A obra descreve os métodos de ionização e a formação dos íons moleculares e quasi-moleculares em cada um desses métodos. O livro tras a interpretação da distribuição de isótopos nas amostras, a identificação dos "parent ions", "base piks" e a distribuição das cargas durante a interpretação dos espectros. A interpretação de sequências de aminoácidos durante a análise de peptídios é rica em dicas, tabelas e exercícios resolvidos. De forma clara é possível entender a lógica por tras da formação das séries de íons A, B, C, X, Y e Z tão importantes durante a identificação da estrutura primária dos peptídeos. Se você pretende trabalhar com a identidficação de compostos de origem protéica utilizando espectrometria de massas, esse deverá ser seu livro de cabeceira!
Broad, thorough introduction.......2003-11-04
I'm not a bench chemist, but I needed a quick survey of how mass spectroscopy is used in handling proteins and other big biomolecules. This book was it.
Although brief, it is thorough and well-organized. The first two chapters are mostly an introduction. Chapter 1 states the problem being solved. The next chapter briefly introduces older technologies, including chemical techiques and 60s-80s mass spec technique. The next two chapters summarize modern mass spec hardware, then start to show how proteins behave in the environment inside the instrument. That gives the fundamentals of protein sequencing: how the molecules break down, and how the fragments help recreate the molecule. The authors go through a few examples in detail, starting from a mass spectrogram and moving forward to sequence. I was especially impressed by the examples that fail. Mass spec analysis is not a magic wand for producing sequences, it is a deductive process, and can not complete an analysis when clues are missing or ambiguous.
The next three chapters are not about mass spec directly. Instead, they discuss how samples are prepared for analysis. This includes the clearest, most informative description of gel electrophoresis that I've seen, along with features of gel chemistry that do or do not interfere with mass spec measurements. This includes a discussion of protein digests, enzymatically produced fragments, and their place in analysis. I would have liked a little more discussion about combining information from digests produced by different enzymes, but no book can cover everything.
The last three chapters extend the discussion of analysis, working upwards from fragments to complete protein sequences. The three chapters respectively address three topics: using standard internet databases for recognizing fragments of known proteins, using combinations of strategies to analyze novel proteins, and using mass spec to identify post-translational modifications. That last one suffers from brevity; perhaps it was only meant to define a problem that deserves a whole book of its own.
Despite its throughness, the authors resist the urge for boggling detail. They present detail up to the point needed for understanding the mechanism and meaning of their topics, then stop. Lots of other writing would benefit from that kind of restraint.
I came away from this book well-informed, and ready to address specific topics in greater detail. That was exactly what I wanted. I recommend this book very highly.

Michael Kinter has presented the topic in a scientific yet enjoyable format. I found the information to be extremely interesting and beneficial in my laboratory. Lets face it folks this isn't the easiest nor most interesting topic to write about. Job well done DR. Kinter. I only have one criticism, there should have been more photos and illustrations. Get your copies quick this will no doubt be on the NewYork times best seller list before long.

Acrylamide levels over a wide range of different food products were analysed using both liquid chromatography-tandem mass spectrometry (HPLC-MS-MS) and gas chromatography-tandem mass spectrometry (GC-MS-MS). Two different sample preparation methods for HPLC-MS-MS analysis were developed and optimised with respect to a high sample throughput on the one hand, and a robust and reliable analysis of difficult matrices on the other hand. The first method is applicable to various foods like potato chips, French fries, cereals, bread, and roasted coffee, allowing the analysis of up to 60 samples per technician and day. The second preparation method is not as simple and fast but enables analysis of difficult matrices like cacao, soluble coffee, molasses, or malt. In addition, this method produces extracts which are also well suited for GC-MS-MS analysis. GC-MS-MS has proven to be a sensitive and selective method offering two transitions for acrylamide even at low levels up to 1@mgkg^-^1. For the respective methods the repeatability (n=10), given as coefficient of variation, ranged from 3% (acrylamide content of 550@mgkg^-^1) to 12% (acrylamide content of 8@mgkg^-^1) depending on the food matrix. The repeatability (n=3) for different food samples spiked with acrylamide (5-1500@mgkg^-^1) ranged from 1 to 20% depending on the spiking level and the food matrix. The limit of quantification (referred to a signal-to-noise ratio of 9:1) was 30@mgkg^-^1 for HPLC-MS-MS and 5@mgkg^-^1 for GC-MS-MS. It could be demonstrated that measurement uncertainties were not only a result of analytical variability but also of inhomogeneity and stability of the acrylamide in food.

The avermectins-ivermectin (IVR), doramectin (DOR), eprinomectin (EPR)-and also the milbemycin moxidectin (MOX) are anthelmintic compounds that may be administered to cattle. Different ionization techniques, including atmospheric pressure photoionization (APPI), were evaluated for the detection of these residues in milk. The ionization response of these compounds using APPI was compared with that obtained by atmospheric pressure chemical ionization (APCI), a combination of APPI and APCI, and electrospray. It was found that the relative response of these drugs with different ionization protocols varied depending on the compound and the mobile phase. When monitoring negative ions, the use of the UV lamp increased the MS response. However, the best response for these compounds was obtained by operating the APCI/APPI source in the positive ion mode without any discharge current applied to the corona needle, whether the UV lamp was on or not. Using this mode of ionization, an MS-MS method was established to monitor the product ion scans of the sodiated molecular ions with an ion trap instrument. Milk fortified with these compounds (0.5-20ngg^-^1) and milk samples from dosed animals were analyzed after isolating the residues with a simple solid phase extraction method. EPR, DOR and IVR were confirmed in all of the extracts analyzed. MOX was confirmed in all samples fortified at 5ngg^-^1 or higher. Acceptable recoveries (>=60%) and relative standard deviations (R.S.D. values @? 20%) were observed for the residues at the following levels: EPR and IVR (1-20ngg^-^1); DOR and MOX (5-20ngg^-^1).

A selective and sensitive liquid chromatography tandem mass spectrometric (LC-MS/MS) method has been developed for simultaneous determination of residual bacitracin and colistin in bovine milk samples. Milk samples were deproteinized and extracted with a mixture of trichloroacetic/formic acid. After centrifugation, bacitracin and colistin in the extracts were separated on a reversed phase Alltima BDS C"1"8 column (250mmx2.1mm, 5@mm) using a gradient elution programme of ammonium formate buffer (saturated ammonium formate:formic acid:acetonitrile:water, 1:5:50:950, v/v/v/v) and 0.1% formic acid in acetonitrile at 0.2mlmin^-^1. Using electrospray LC-MS/MS with time-scheduled multiple reaction monitoring (MRM), identification and quantification of the major components of the two polypeptides were performed based upon the intensities of mass fragments from the respective doubly charged precursor ions: bacitracin A at 712->199amu and 712->227amu, and colistin A at 586->101amu, 586->202amu and 586->241amu, respectively, at the defined retention time windows and the matching of the specific tolerance of the relative abundance of the monitored ions. Polymyxin B, a polypeptide of colistin family, was employed as the internal standard. With an additional solid phase extraction procedure, the analytical method was applicable to the analysis of the two polypeptide antibiotics in animal's liver and tissue samples. The practical quantification limits of bacitracin A and colistin A were 100 and 50@mgkg^-^1; and mean intra-day (n=7) and inter-day (n=4) recoveries were in the range of 89.2-110% (R.S.D.
<10%), and 90.4-112% (R.S.D.
<13%) respectively in all matrices.

Recent reports from the Brazilian Agricultural Ministry program of residue control describe some cases of the presence of semicarbazide, the target metabolite of nitrofurazone {2-[(5-nitro-2-furanyl)methylene]-hydrazinecarboxamide}, in samples of salted chicken meat. Since that contamination can be originated from other reasons in addition to nitrofurazone abuse, our laboratory has developed a sensitive and simple method for detection of four nitrofuran metabolites in salt samples. This method include a previous hexane extraction of the metabolites followed by a derivatization reaction with 2-nitrobenzaldehyde. The nitrophenyl derivatives were analyzed by liquid chromatography-tandem mass spectrometry without any additional clean-up procedures. The limit of quantification for routine analysis was 0.5@mgkg^-^1 for each metabolite.

A new IC-ESI-MS/MS method, with simple sample preparation procedure, has been developed for quantification and confirmation of perchlorate (ClO"4^-) anions in water, fresh and canned food, wine and beer samples at low part-per-trillion (ngl^-^1) levels. To the best of our knowledge, this is the first time an analytical method is used for determination of perchlorate in wine and beer samples. The IC-ESI-MS/MS instrumentation consisted of an ICS-2500 ion chromatography (IC) system coupled to either an API 2000(TM) or an API 3200(TM) mass spectrometer. The IC-ESI-MS/MS system was optimized to monitor two pairs of precursor and fragment ion transitions, i.e., multiple reaction monitoring (MRM). All samples had oxygen-18 isotope labeled perchlorate internal standard (ISTD) added prior to extraction. Chlorine isotope ratio (^3^5Cl/^3^7Cl) was used as a confirmation tool. The transition of ^3^5Cl^1^6O"4^- (m/z 98.9) into ^3^5Cl^1^6O"3^- (m/z 82.9) was monitored for quantifying the main analyte; the transition of ^3^7Cl^1^6O"4^- (m/z 100.9) into ^3^7Cl^1^6O"3^- (m/z 84.9) was monitored for examining a proper isotopic abundance ratio of ^3^5Cl/^3^7Cl; and the transition of ^3^5Cl^1^8O"4^- (m/z 107.0) into ^3^5Cl^1^8O"3^- (m/z 89.0) was monitored for quantifying the internal standard. The minimum detection limit (MDL) for this method in de-ionized water is 5ngl^-^1 (ppt) using the API 2000(TM) mass spectrometer and 0.5ngl^-^1 using the API 3200(TM) mass spectrometer. Over 350 food and beverage samples were analyzed mostly in triplicate. Except for four, all samples were found to contain measurable amounts of perchlorate. The levels found ranged from 5ngl^-^1 to 463.5+/-6.36@mgkg^-^1 using MRM 98.9->82.9 and 100@ml injection.

I read this book about 10 years ago......and today I'm still laughing.
Jerome possessed very good comic timing, the story flowed naturally, the emotions and reaction of the 3 men were so real that I wish I was on the river and met them at that time! Somehow this book can teach us the importance of taking a break from our daily life.
I recently bought a copy from Amazon which was initialy meant as a gift for someone, but having a second thought, I decided to keep it as a precious reading companion for myself when I'm travelling.

Jerome K. Jerome might have been a riot 100 years ago but this book will provoke only a rare smirk. The best bits belong to the dog who only seems to appear at the end of every third or fourth chapter to drop a one-liner. Other than that, it is aged, slow moving slapstick with some oddly out-of-place moments of reflection.
How anyone can call this a laugh riot boggles my mind.

I had put off reading this book for many years, and finally got to it.
The writing in this novel is simply superb. Jerome possesses a comic timing that most authors don't have and can simply never hope to have. He is able to weave in a history of the River Thames in with the bumblings of the three main characters and their dog, without hurting either major premise of the book.
The main reason to read this is because it will make you laugh. If it doesn't, you are not understanding it correctly.

Imagine Bertie Wooster and two of his idiot friends out on a boat... with no Jeeves. That about describes "Three Men in a Boat : To Say Nothing of the Dog," Jerome K. Jerome's enchanting comic novel about three young men (to say nothing of the dog) who discover the "joys" of roughing it.
The three men are George, Harris and the narrator, who are all massive hypochiandriacs -- they find that they have symptoms of every disease in existance (except housemaid's knee). To prop up their failing health, they decide to take a cruise down the Thames in a rented boat, camping and enjoying nature's bounty.
Along with Monty -- an angelic-looking, devilish terrier -- the three friends set off down the river. But they find that not everything is as easy as they expected. They get lost in hedge mazes, end up going downstream without a paddle, encounter monstrous cats and vicious swans, have picnics navigate locks, offend German professors, and generally get into every kind of trouble they possibly can...
Even though it was published more than a century ago, "Three Men in a Boat" remains as freshly humorous as when it was first published. While editor/playwright/author Jerome K. Jerome wrote a lot of other books, this book remains his most famous. And once you've read it, you'll see why.
Jerome's real talent is in finding humor in everyday things, like trying to erect a tent in the woods, getting seasick, or questioning whether it's safe to drink river water. Written in Jerome's dry, goofy prose, these little occurrances become immensely funny. One of the funniest parts of the book is when the boys listen to a fishermen telling of his prowess, only to accidently knock down his record-breaking stuffed fish.... and discover it's made out of plaster. Oops.
But Jerome takes a break from the humor near the end, when the boys find a drowned woman floating in the river. And here he becomes solemn and quietly compassionate: "She had sinned - some of us do now and then - and her family and friends, naturally shocked and indignant, had closed their doors against her."
But back on the funny stuff. The capstone on all this humor is the "three men." These guys are basically pampered Victorian aristocrats, who have a romantic yearning for the great outdoors. You'll be laughing at them and with them, as they struggle through the basics of boating and camping.
It's worth noting that the Digireads edition of this book is very good, with a flexible cover, extremely strong binding, and a nice reproduction with rather small print. Think "Dover Thrift," but of higher quality.
Funny, wacky and creepily true to life, "Three Men in a Boat" is an enduring comic classic in the vein of PG Wodehouse. Not to mention the dog!
