    Book Description

X-ray crystallography has long been a vital method for studying the structure of proteins and other macromolecules. As the importance of proteins continues to grow, in fields from biochemistry and biophysics to pharmaceutical development and biotechnology, many researchers have found that a knowledge of X-ray diffraction is an indispensable tool. In this new edition of his essential work, Dr. Jan Drenth, recognized internationally for his numerous contributions to crystallographic research, has provided an up-to-date and technically rigorous introduction to the subject.

Principles of Protein X-ray Crystallography provides the theoretical background necessary to understand how the structure of proteins is determined at atomic resolution. It is intended to serve as an introduction for graduate students, postdoctoral researchers, and established scientists who want to use protein crystallography in their own endeavors, or need to understand the subject in order to critically evaluate the literature. New additions to the book include a section on twinning, an additional chapter on crystal growth and a discussion of single-wavelength anomalous dispersion (SAD).

    About the Authors:

Dr. Jan Drenth is a professor emeritus at the Laboratory of Biophysical Chemistry at the University of Groningen, The Netherlands.

Contributing author Dr. Jeroen R. Mesters Ph.D. is a Senior Research Assistant at the Institute of Biochemistry, University of Luebeck, Germany.

4 out of 5 stars Depends on who you are and what you want..........2007-09-28

Jan Drenth is an old school protein crystallographer who has seen the evolution of the field right from its inception in the 1950s to the present day. He belongs to the era when Protein Crystallography was dominated by people with serious math and physics skills like Maurice Wilkins, Rosalind Franklin, Francis Crick, Max Perutz and John Kendrew. Owing to the lack of good computers in those days, all the calculations had to be done by hand and there was no way you could solve a protein structure if you didn't know what a Fourier Transform was. The situation is very different nowadays where, thanks to computers, even a person who doesn't know how to differentiate a function can solve protein structures.
Understandably, owing to the author's experiences and background, this book is an excellent one for people interested in the real math behind protein crystallography. It is much on the lines of the other classic book by Blundell and Johnson, also crystallographers belonging to the same era as Jan Drenth. It is quite full of mathematics and therefore, is not recommended for biologists or chemists with weak math skills. For such people I would recommend other introductory books on the subject written by Gale Rhodes, David Blow and Alex McPherson. A good approach would be to learn the basics from these intoductory books and 'graduate' to Drenth's book later.

1 out of 5 stars Not Helpful for a introductory class.......2006-12-07

I used this text book for an introductory graduate class in X-ray Crystallograhy. After taking a semester of X-ray Crystallograhy (which is labeled as a Chemistry and Biochemistry course), I can say that the material is mainly based on physics and mathematics. I can't say that this text helped a great deal for my course. The text does not explain relevant points such as Miller indices, symmetry elements, or space groups clearly or in much depth. Instead, it spends too much time explaining concepts mathematically and deriving equations. It also does not contain any relevant practice problems, which are needed to understand Crystallography and practice for exams.

X-ray Crystallography is a very dense subject, and is not a course to take on a whim. I would not reccomend taking a course on Crystallography unless you have a really intense interest in pursuing research in that area. If you are interested in this area, do not buy this text even if it is required for your course. Take a look at Crystallography Made Crystal Clear by Gale Rhodes. That book is much clearer, though also lacks sufficient practice problems.

3 out of 5 stars Decent text, but better Crystallography books are out there........2002-09-16

Drenth is a decent enough book. The figures are ok, nothing great. They mostly just drive home the point made in the text, but are not instructive in and upon themselves. If you are looking to get a basic understanding of X-Ray crystallography and its uses to solve protein structure try the Gale Rhodes book. If you are looking for a more rigorous book try the Michael M. Woolfson book. Drenth tries to be the middle ground between the two. The subject though is too complicated to do it justice like that. Either keep it simple or go in depth.

5 out of 5 stars Best introductory text.......2001-04-17

Best introductory text for macromolecular xray crystallography. Simple and succinct mathematical explanation of basic concepts. It provides enough background for students to start a research project.

5 out of 5 stars A great introduction in crystal structure analysis.......2004-01-29

This book should be one of the best introduction in crystal structure analysis for the beginners! Everybody wanting to start with x-ray structure analysis should read this book at the very beginning, since it's, in fact, a International Union of Crystallography Text on Crystallography. It covers the most important topics of x-ray structure analysis in an easy understandable language without boring the reader with mathematical details. You can get a good impression about what the basic principles and problems of x-crystallography are, and how they can be solved.

      Book Description

Quantum Chemistry of Solids delivers a comprehensive account of the main features and possibilities of LCAO methods for the first principles calculations of electronic structure of periodic systems. The first part describes the basic theory underlying the LCAO methods applied to periodic systems and the use of wave-function-based (Hartree-Fock), density-based (DFT) and hybrid hamiltonians. The translation and site symmetry consideration is included to establish connection between k-space solid-state physics and real-space quantum chemistry methods in the framework of cyclic model of an infinite crystal. The inclusion of electron correlation effects for periodic systems is considered on the basis of localized crystalline orbitals. The possibilities of LCAO methods for chemical bonding analysis in periodic systems are discussed. The second part deals with the applications of LCAO methods for calculations of bulk crystal properties, including magnetic ordering and crystal structure optimization. The discussion of the results of some supercell calculations of point defects in non-metallic solids and of the crystalline surfaces electronic structure illustrates the efficiency of LCAO method for solids.

              Book Description

Recently, major progress has been made in the development of equations to describe the motion of fluid-particle mixtures and their application to a limited range of problems. However, results are only as good as their underlying equations, so it is essential to have a clear understanding of the fundamental physics. In The Dynamics of Fluidized Particles, Jackson formulates these equations carefully and fully describes the important existing applications that test their ability to predict salient phenomena.

                Book Description

This book provides a much needed and thorough treatment of the heat transfer in agitated disperse systems. It gives predictive equations for the heat transfer in moving beds, bubbling and circulating fluidized beds, pneumatic transport in vertical tubes and particulate fluidized beds. Owing to the many different modes of activation of heat transfer, the basic approach of the book is to provide experimental evidence of the relevance of particle motion to the proximity of solid surfaces for the heat transfer observed. This has been achieved by the evaluation of experiments obtained with a newly developed pulsed light method using luminous particles. Heat Transfer in Fluidized Beds will be of great use to students and researchers involved in heat transfer and thermodynamics.

                  Description:
Fluidized bed filtration classified as granular bed filtration is a solution for removing particles from flue gas. Fine particles are captured by colliding with bed material. Binary mixtures including fines exist in the fluidized bed; the situation becomes more complex because the fines increases with time as particles enter continuously. Coal ash was put into the inlet gas of a gas-solid fluidized bed with silica sand and the variations in concentration with time was monitored, to elucidate the dynamic purification of fines by bed material and the interaction therewith. Water was injected into the inlet gas also to study the influence of water content on the removal of particles. Experimental results showed that the particle concentration rapidly increased with time until equilibrium was achieved. The outlet concentrations exceeded the inlet concentration (such that the removal efficiency was negative) at operating time between 26 and 30min, yielding an efficiency as low as -35%. The removal efficiency increased with the relative humidity from 30% to 70%. The maximum cumulative collection ratios of particles (by mass) were 20%, 22% and 27% at humidities of 30%, 50% and 70%, declining to 7%, 13%, and 19% at the end.

                    Description:
The dynamics and structure of a fluidized bed in inclined columns are investigated experimentally. Experiments were conducted in two glass columns of L/D=95 and 150, and diameters of 2.58 and 2.78cm, which can be positioned in the whole range of inclination angles, from horizontal to vertical. The fluidizing agent is water and the grain material is particles of different densities and characteristic diameters. Measurements of the pressure loss, flow rate of the fluidizing agent and fluidized bed position and height are carried out. Particle flow patterns were visualized using a digital camera. The expansion process of the fluidized bed depends strongly on the inclination angle. Consequently the critical velocity for the bed escape varies with the column inclination angle and exhibits a maximum at about 45^o. The column length was found to have a minor effect on the phenomena involved.

                      Description:
A three-dimensional particle dynamic analyzer (3D-PDA) was employed to measure the gas-solid two-phase flow in a lab-scale square cyclone separator with downward gas-exit. Several cases of different inlet velocity and particle concentration were studied. The particle used in the test was glass bead of mean diameter 30-40@mm. Discussion was given on the distribution of flow vector, mean velocity, turbulent intensity and kinetic energy of both gas and particles of different diameter at different position in the separator. It was found that the center of the flow field deviated from the geometric center of the cyclone. The flow fields had the feature of Rankine eddy, i.e., strongly swirling region in the central part and pseudo-free eddy region of weak swirling intensity near the cyclone wall. Local vortex existed at the corners where the flow changed its direction sharply. When the cyclone wall was heated and the suspension temperature was elevated, the flow field became more uniform than that under room-temperature condition. The local vortexes at the corners were weakened and the swirling intensity became poorer which led to decreased total mean separation efficiency from about 81% to 76.5%. The right-side wall facing the suspension inlet gave the major contribution to the separation efficiency, where the largest downward velocity was measured. Back-flow (upward velocity) was found around the center of the separator above the gas-exit. The quasi-laminar motion of particles enhanced the turbulent motion at the corners due to particle-particle or particle-wall collision, which led to the local peak value of the turbulent kinetic energy and turbulent intensity. The corner was one of the major regions to cause pressure drop and was found to be beneficial to particle separation mainly because the strong fluctuating flow consumed much of the kinetic energy of both the particle and gas.

The ensuing day was spent, partly in sleep, and partly in languor and disquietude. I incessantly ruminated on the incidents of the last night. The scheme that I had formed was defeated. Was it likely that this unknown person would repeat his midnight visits to the Elm? If he did, and could again be discovered, should I resolve to undertake a new pursuit, which might terminate abortively, or in some signal disaster? But what proof had I that the same rout would be taken, and that he would again inter himself alive in the same spot?

3 out of 5 stars Early American Romance.......2003-04-19

Often ranked as "the first significant American novelist"-this is how Norman Grabo characterizes him in the Introduction to this volume-Charles Brockden Brown was an ambitious and inventive teller of tales, although an awkward literary craftsman. Brown was only in his twenties when he published this novel in 1799, but it was already his fourth book. Edgar Huntly, which takes place in rural Pennsylvania in 1787 recounts the strange adventures of a young man who sets out to discover the person responsible for killing his best friend, Waldegrave, who has recently died under mysterious circumstances. His investigations put him on the track of Clithero, an Irish servant employed in his uncle's household, but one thing leads to another and Edgar finds himself having to fight Indians and face the perils of the wilderness in order to make his way back home. Most of the story is told by Edgar himself in a long letter-some twenty-seven chapters long-that he is in the process of writing to his intended, Waldegrave's sister, Mary.
Edgar Huntly belongs to the genre of romance, the much older but somewhat less respectable sibling of the novel of social realism that had come into vogue in the eighteenth century. The romance frequently has an exotic setting, and features incidents that stretch the limits of artistic plausibility, where it does not take a plunge into fantasy, as it does in Matthew Gregory Lewis's The Monk or Charles Robert Maturin's Melmoth the Wanderer. Nevertheless, the genre enjoyed great popularity here down to the time of the Civil War, and Brown shows himself well acquainted with its conventions. He not only throws in a whole series of hair-raising encounters that pit the inexperienced Edgar with natural hazards, predatory wild animals, and marauding Delawares, but supplies a convoluted plot line that he further complicates with stories-within-the main story told by subordinate characters. Even for a romance, Edgar Huntly has an unusually tangled narrative web. It's hardly surprising the neophyte author himself sometimes has difficulty keeping track of the strands.
The reader making the acquaintance of Brown for the first time will not get any help from the note on the back cover supplied by Penguin, according to which "Edgar Huntly is the story of a young man who sleepwalks each night, a threat to himself and others, unable to control his baser passions....One of America's first Gothic novels...." I wonder whether the person responsible for these inane comments ever bothered to open the book. In the first place, Edgar Huntly is no Gothic novel. As E.F. Bleiler pointed out, it takes a castle to make a Gothic novel. But Brown explicitly distances himself from the suspicion of Gothicism in the remarkable address "To the Public" prefaced to the book, in which he prides himself on having found his materials in his native country and rejoices in not having fallen back on "Gothic castles and chimeras" in composing his work. But the statement about Edgar is not just inaccurate-it is blatantly incorrect. Edgar has at the most two sleepwalking episodes, one of which serves to initiate the most remarkable series of events in the novel, when he awakes to find himself mysteriously transported to a cave in the middle of the night. And nothing Edgar relates suggests he has a history of somnambulism in his past-nor that he is "unable to control his baser passions." In fact, the first sleepwalker to show up is the far more uncontrolled Clithero, who almost seems to have infected Edgar with his affliction.
Brown was clearly a pioneer of psychological analysis in the history of the novel. Like Edgar Allan Poe later, he probed the souls of his characters by plunging them into violent, imminently lethal situations. As a student of extreme states of the human psyche, he was not only a predecessor of Poe, but of Hawthorne and Melville as well. Yet Brown lacked the ability to apply his talent to the creation of highly individualized characters, one of the strengths of great nineteenth century novelists such as Charles Dickens, Gustave Flaubert, and Fyodor Dostoevsky. All of the characters in Edgar Huntly, the protagonist included, remain little more than phantoms inhabiting a largely crepuscular world throughout the course of the action. However, like other trailblazing figures in the early history of American fiction-James Fenimore Cooper is a perfect example-Brown had an estimable ability to create atmosphere. It is not intended as a sarcasm to say that the reader may feel he or she is turning into a sleepwalker while reading Edgar Huntly.

5 out of 5 stars Another masterpiece overlooked..........2002-03-07

This book is truly one of the most interesting and purely entertaining books I've ever read. The creepy imagery of the sleepwalking man digging in the street and then the caves and indians will stick with me forever. What an amazingly creative man. An x-files-esque dreamer living in the 1790s.

5 out of 5 stars What Lies Beneath.......2001-08-24

First published in 1799, Charles Brockden Brown's novel, "Edgar Huntly" is an insane masterpiece. I love Brown. I read this novel for the first time a few years ago, and thought it was about time to refresh my memory. Set outside of Philadelphia in the final years of the 1780's, "Edgar Huntly," like other of Brown's works, test the new American republic's capacity to govern while balancing the needs and desires of its culturally disparate inhabitants.

The novel is purportedly a correspondence from the protagonist, Edgar Huntly, to his friend/love interest, Mary Waldegrave, in the aftermath of her brother's death. Edgar is an educated, refined, enlightened young man, disconsolate upon the death of his friend. An avid walker, Edgar frequently leaves the environs of his hometown, Solebury, returning to the scene of his friend's death, a large elm tree. Near this tree late one evening, he spots a man, conspicuously lurking, burying something beneath the tree. Suspecting this man, Clithero, of Waldegrave's murder, Edgar begins a career of surveillance and tracking, following Clithero to his residence and through the uncharted wildernesses that border his hometown. What follows is Edgar's progress in discovering the truths behind the death of Waldegrave, the history of Clithero, and the foundations of his own self-control and rationality.

Brown deals with a number of issues throughout the novel current to late 18th century America, including the dispossession of Native Americans from their land, Irish immigration, and the instability of a newly formed nation. Philosophically, Brown examines popular 18th century debates over the limits of sympathy, and the ability of sense, experiment, and observation to conclusively explain human nature. In his preface to the novel, Brown says that his novel will not exploit the then-common motifs of gothic fiction. Perhaps, but Brown, taking the example of William Godwin, moves the castles, dungeons, and murders of traditional gothic into the psyches of his characters. Dementia, paranoia, and in this novel, at least, the uncontrollability of sleep-walking, constitute the largely internal threats to personal and national safety.

So join Edgar, Clithero, Sarsefield, the Lorimers, Inglefield, Queen Mab, and an army of hostile natives, on an intricate, often horrifying romp through late 18th century America. Brown's doubts and fears about living in the new nation will entrance and mortify you, and possibly make you consider putting yourself in restraints before you go to bed at night.

                          4 out of 5 stars Brockden Brown's Twisted Imagination.......2001-04-03

This novel is an excellent journey through the dark, disturbing wilderness of 18th century America and its dangers. The wars that raged between Natives and their oppressors (the English) are here brutally portrayed. With a new introduction to English literature of the effects of sleepwalking, this is also a journey through the psychology of a psychotic sleepwalker, and the devils that haunt him. An excellent read for those searching for the natural horror of humanity, with a little suggestion of the supernatural, though very heavy both in language, and symbolism and metaphor. A tough read, but well worth it.

4 out of 5 stars Very Interestingly Confusing.......2000-12-03

This book (the cover amazon has displayed is *wrong*) is very interesting. It is also exceedingly strange. It was never boring, but it never made too much sense either. You think there might be supernatural elements involved -- but maybe not. It's a tale of the coming to America from England, the stuff that happens to the British here, and Indians. Cool, confusing, book.
